MicroRNA-33 Inhibits Adaptive Thermogenesis and Adipose Tissue Beiging

Milessa Silva Afonso, Narendra Verma, Coen van Solingen, Yannick Cyr, Monika Sharma, Luce Perie, Emma M. Corr, Martin Schlegel, Lianne C. Shanley, Daniel Peled, Jenny Y. Yoo, Ann Marie Schmidt, Elisabetta Mueller, Kathryn J. Moore

Summary: Recent studies have shown that miR-33 plays a crucial role in regulating metabolic pathways in brown adipocytes, with reduced levels observed in response to thermogenic activators. In mice exposed to cold, miR-33 levels in BAT are rapidly downregulated, suggesting a role in repression of adaptive thermogenesis. Inhibition of miR-33 in mice leads to increased expression of key genes involved in BAT activation and white adipose beiging, rendering mice resistant to cold challenge.

miR-33 Silencing Reprograms the Immune Cell Landscape in Atherosclerotic Plaques

Milessa Silva Afonso, Monika Sharma, Martin Schlegel, Coen van Solingen, Graeme J. Koelwyn, Lianne C. Shanley, Lauren Beckett, Daniel Peled, Karishma Rahman, Chiara Giannarelli, Huilin Li, Emily J. Brown, Alireza Khodadadi-Jamayran, Edward A. Fisher, Kathryn J. Moore

Summary: Inhibition of miR-33 shifts the balance of proatherosclerotic and antiatherosclerotic immune cell subsets in atherosclerotic plaques, reducing plaque burden and inflammation. This treatment also affects the expression of specific target genes in different macrophage subsets, ultimately leading to a decrease in aortic CD8+ T cells and CD4+ T-helper 1 cells, and an increase in regulatory T cells within plaques. These findings shed light on the immune mechanisms underlying the atheroprotective effects of anti-miR-33.

Summary: Targeting Ntn1 in advanced atherosclerosis can ameliorate atherosclerotic inflammation and promote plaque regression by restructuring monocyte-macrophage dynamics and enhancing pathways involved in inflammation resolution.

High-Throughput Screening Identifies MicroRNAs Regulating Human PCSK9 and Hepatic Low-Density Lipoprotein Receptor Expression

Coen van Solingen, Scott R. Oldebeken, Alessandro G. Salerno, Amarylis C. B. A. Wanschel, Kathryn J. Moore

Summary: The discovery of PCSK9 and its role in LDL regulation has led to the rapid development of therapeutic PCSK9 inhibitors. Research has identified certain microRNAs that can reduce PCSK9 expression, leading to increased LDLR expression in hepatic cells, which may have therapeutic implications for manipulating LDLR expression and cholesterol levels in the liver.
