Summary: MicroRNAs (miRNAs) are small non-coding RNAs that play a role in regulating pathophysiological processes by suppressing target gene expression post-transcriptionally. The function of miRNAs is regulated by a conserved biogenesis machinery involving processing steps and the formation of a miRNA-induced silencing complex (miRISC). This study discovered a novel regulatory mechanism involving the post-translational modification of AGO2 by E-cadherin, leading to enhanced miRISC activity on gene silencing.

Summary: Asymmetric stem cell division (ASCD) is a crucial mechanism in development, cancer, and stem cell biology. In this study, the researchers show that the extrinsic axon guidance cues Netrins regulate NB ASCD in Drosophila larval brain neural stem cell lineages. They demonstrate that the signaling pathway involving Netrin-Frazzled/DCC and Abelson kinase modulates Robo1 signaling levels, which affects the process of asymmetric stem cell division. Mechanistically, Robo1 signaling directly interacts with several key components of the intrinsic ASCD machinery, ultimately leading to the generation of ectopic NBs and progenitor cells.

Summary: The PML protein forms aggregates called PML nuclear bodies (PML-NBs) in the nucleus, where it regulates the localization of transcription factors. It is also associated with chromatin in different cell types, indicating its role in transcriptional regulation. In this study, we investigated the association of PML with chromatin in triple-negative breast cancer (TNBC) cell lines, where it plays a critical oncogenic role. We found that PML associates discontinuously with large heterochromatic domains, known as PML-associated domains (PADs), which contain gene-rich sub-domains. PML promotes heterochromatic organization and expression of pro-metastatic genes within these sub-domains. Importantly, this regulation occurs outside PML-NBs, suggesting that PML has a relevant gene regulatory function in the nucleoplasm. We also found that PML indirectly regulates pro-metastatic genes outside PADs in TNBC cells. Our findings highlight the importance of PML as a transcriptional regulator in TNBC cells, contributing to the expression and organization of pro-oncogenic genes in specific chromatin domains.
