Summary: This passage discusses the utility of dinucleotide analogs of messenger RNA cap, particularly those with benzyl substitution at N7 position, for potential translational inhibition. The synthesis of novel Bn(7)GpppG analogs and their evaluation in terms of translation and translational inhibition are highlighted, showing promising results for N7-(4-halogenbenzyl) substitutions. Additionally, crystal structures of compounds with eIF4E were determined to gain more molecular insight into N7-benzylation of mRNA cap.

Summary: Among CNS cancers, glioblastoma (GB) is the most common and aggressive type. Current treatments involve surgery, radiation, and chemotherapy, but the prognosis remains poor. Recent studies have focused on mRNA translation and its dysregulation in GB, particularly in the initiation phase. The translation machinery and ribosomal proteins have also been implicated in GB development. This review discusses the relationship between translation initiation, the translation machinery, and GB, as well as the potential of targeting the translation machinery for improved patient survival.

Summary: Hypoxia causes changes in gene expression in eukaryotic cells by reducing ATP supply and inhibiting protein synthesis. This study shows that a CA-rich motif in the 3'untranslated region of Ldh mRNA allows it to be highly translated in hypoxia. The cap-binding protein eIF4EHP is identified as a key factor in 3'UTR-dependent translation under low oxygen conditions, and it is also necessary for Drosophila development and mobility after hypoxic challenge.

The results showed efficient phosphorylation of 4E-BP1 at lysosomes and the plasma membrane, while it was decreased in the nucleus. Summary: Many eukaryotic RNA viruses regulate gene expression through transcription of subgenomic mRNA, which is controlled by intragenomic interactions forming higher-order RNA structures. However, a new study reveals that an umbravirus activates sg mRNA transcription through base pair-mediated dimerization of its RNA genome. The study provides evidence of a kissing-loop interaction that forms a dimer,located just upstream of the transcriptional initiation site, and is involved in transcriptional activation. This unconventional transcription strategy may be utilized by other similar viruses.

Summary: The lifecycle of RNA polymerase II (RNAPII) transcripts is influenced by the formation of mutually exclusive ribonucleoprotein complexes (RNPs) that guide transcript generation and degradation. A recent study reveals that the scaffold protein ARS2, bound to the cap binding complex (CBC), regulates RNA metabolism in the nucleus. Interestingly, alternative splicing of ARS2 generates cytoplasmic isoforms that exhibit different functions from their nuclear counterparts, suggesting their importance in cellular homeostasis and stress response.

Summary: Translation in pathogenic protozoans of the family Trypanosomatidae involves multiple eIF4F-like complexes, with the EIF4E4/EIF4G3 and EIF4E3/EIF4G4 pairs being the most prominent. These complexes exhibit distinct protein partners and play important roles in gene expression and cell morphology. Knockdown studies revealed that EIF4E3/EIF4G4 are essential for viability and are involved in cytokinesis. The differential association of mRNAs with each complex was also identified through CLIP-seq analysis, further confirming their distinct functions.

Summary: Increased expression of the peptide hormone retinol-binding protein 4 (RBP4) has been linked to insulin resistance and type 2 diabetes. Studies have shown that translational control of RBP4 synthesis plays a role in its regulation, with activation of mTORC1 kinase promoting hepatic Rbp4 mRNA translation. Targeting Rbp4 mRNA translation could be an alternative therapeutic approach to reduce serum RBP4 levels.
