Genetic variability of the stolbur phytoplasma vmp1 gene in grapevines, bindweeds and vegetables

Aim: Evaluation of the genetic variability of stolbur phytoplasma infecting grapevines, bindweeds and vegetables, collected in different central and southern Italian regions. Materials and Results: Phytoplasma isolates belonging to stolbur subgroup 16SrXII-A were subjected to molecular characterization by polymerase chain reaction/restriction fragment length polymorphism (PCR/RFLP), to investigate two different nonribosomal genes: tuf and vmp1. In grapevines, 32% of samples were infected by tuf-a type and 68% by tuf-b type, with different relative incidences in the regions surveyed. All herbaceous samples (bindweeds, tomato, tobacco, pepper, celery) were infected by tuf-b. The gene vmp1 showed higher polymorphism in grapevines (nine profiles) than herbaceous plants (six) by RFLP analysis, in agreement with nucleotide sequences' analysis and virtual digestions. Conclusions: The phylogenetic analysis of vmp1 gene sequences supports the RFLP data and demonstrates the accuracy of RFLP for preliminary assessments of genetic diversity of stolbur phytoplasmas and for screening different vmp types. Significance and Impact of the Study: Stolbur represents a serious phyto-sanitary problem in the areas under investigation, owing to heavy economic losses in infected grapevines and vegetables. Molecular information about the complex genotyping of the vmp1 gene provides useful data towards a better understanding of stolbur epidemiology. Moreover, this study clarifies some different vmp1 genotype classifications of stolbur, providing molecular data in comparison with previous investigations.

Epiphytic Yeasts and Bacteria as Candidate Biocontrol Agents of Green and Blue Molds of Citrus Fruits

Summary: In this study, yeast and bacterial isolates with antagonistic activity against Penicillium digitatum and P. italicum on citrus fruits were screened and identified. These isolates significantly reduced the incidence of green and blue mold caused by these pathogens on citrus fruits, and effectively prevented fungal infection during low-temperature storage. Moreover, they did not have any negative effect on fruit quality and shelf life. These selected isolates have multiple modes of action and show promise as biocontrol agents for postharvest green and blue molds of citrus fruits.

Summary: Fresh fruits and vegetables continue metabolic activity after harvest, leading to ripening, increased susceptibility to decay-causing fungi, and consequent loss and waste. Edible coatings can reduce waste, respect the environment, and benefit consumer health. They act as a protective agent, extending shelf life, and have the potential to control ripening and maintain nutritional properties of fresh produce.

Chitosan and chitooligosaccharide regulated reactive oxygen species homeostasis at wounds of pear fruit during healing

Summary: Both chitosan (CTS) and chitooligosaccharide (COS) can regulate reactive oxygen species (ROS) homeostasis during wound healing of pear fruit by scavenging excessive H2O2 and improving antioxidant capacity. They increase NADPH oxidase and superoxide dismutase activities, promote O2.- and H2O2 production, enhance the activities of several enzymes, and elevate the levels of ascorbic acid and glutathione. In addition, they improve antioxidant capacity in vitro and maintain cell membrane integrity at fruit wounds. COS demonstrates superior performance over CTS.

Distribution Quality of Agrochemicals for the Revamping of a Sprayer System Based on Lidar Technology and Grapevine Disease Management

Summary: Grapevines are a significant concern due to their potential risk to health and biodiversity. Therefore, it is crucial to control the distribution of agrochemicals to achieve high quality and sustainable viticulture. Field tests using a LIDAR-based prototype coupled with a commercial sprayer showed improved distribution performance and reduced incidences of grapevine diseases. This innovative sprayer, with its on/off control system, can effectively reduce chemical distribution and regulate vineyard fungal diseases, contributing to sustainable and high-quality production.

Hydrolate and EO Application to Reduce Decay of Carica papaya during Storage

Summary: Postharvest fruit loss is caused by the absence of advanced handling and storage technologies and the presence of fungal pathogens. This study focused on the use of edible coatings derived from Origanum vulgare L. subsp. viridulum essential oil and Aloe arborescens Mill. gel to provide defense against fungal infections in papaya. The results showed that fruits coated with the combination of essential oil and gel had higher fungal contamination, while fruits coated with the hydrolate only showed better preservation of organoleptic characteristics with lower decay index. These findings suggest that the use of hydrolate can be a sustainable alternative to synthetic fungicides for extending the shelf life and maintaining the quality of papaya fruit.

The observation of taxonomic boundaries for the 16SrII and 16SrXXV phytoplasmas using genome-based delimitation

Preharvest chitooligosaccharide spray alleviates chilling injury in harvested muskmelon fruit by regulating membrane lipid metabolism and activating antioxidant enzyme activity

Summary: This study found that preharvest chitooligosaccharide (COS) spray can alleviate chilling injury (CI) in muskmelon fruit by regulating membrane lipid metabolism and increasing antioxidant enzyme activity. The COS spray up-regulated the expression of CmPLA(2), promoting the release of unsaturated fatty acids and increasing the double bond index (DBI) value. It also down-regulated the expression of CmPLC and CmPLD, increased the levels of phosphatidylcholine (PC) and phosphatidylinositol (PI), and decreased the levels of phosphatidic acid (PA). Furthermore, the COS spray increased the activity of superoxide dismutase (SOD) and catalase (CAT), decreased O-2(-) and H2O2 levels, maintained cell membrane permeability, and reduced MDA levels in fruit during chilling, thereby alleviating CI.

Effects of Commercial Natural Compounds on Postharvest Decay of Strawberry Fruit

Summary: This study tested the effectiveness of six natural compounds in controlling postharvest decay of strawberries. Chitosan-based coating compounds and citrus essential oil products were effective in controlling gray mold under cold storage conditions, while methyl jasmonate plant growth regulator and grapefruit seed essential oil products were more effective at room temperature. Humic acid and chitosan-based coating compounds were more effective in controlling Rhizopus rot under both cold storage and room temperature conditions.

The relationship between occlusions and esca symptoms was investigated across different countries and cultivars, highlighting the unique impact of occlusions on leaf physiology.
