Mitochondrial sirtuins in stem cells and cancer

Summary: There is a potential relationship between subfertility and peripartum cardiomyopathy (PPCM), with subfertile PPCM patients having a higher risk of cardiovascular complications. However, the use of assisted reproduction technology (ART) does not increase the risk of PPCM. Close monitoring is recommended for subfertile women to detect signs of peripartum heart failure.

Palmitate induces DNA damage and senescence in human adipocytes in vitro that can be alleviated by oleic acid but not inorganic nitrate

Summary: This study successfully corrected a COL17A1 gene mutation associated with junctional epidermolysis bullosa using Cas9 nuclease and nickase-based targeting strategies. The corrected cells restored expression of C17 protein and showed improved adhesion capabilities. This research represents the first gene-editing-based correction of a COL17A1 mutation and demonstrates the superiority of proximal paired nicking strategies based on Cas9 D10A nickase for gene reframing in a clinical context.

Batch Effects during Human Bone Marrow Stromal Cell Propagation Prevail Donor Variation and Culture Duration: Impact on Genotype, Phenotype and Function

Summary: Donor variation is an important issue limiting cell-based therapies. The use of human platelet lysate (hPL) instead of fetal bovine serum (FBS) to propagate bone marrow stromal cells (BMSCs) can have significant effects on BMSC phenotype and function. Notably, propagation in hPL increased BMSC proliferation and gene expression trajectories, resulting in distinct surface marker signatures after just one passage. The use of FBS as a culture medium led to declining proliferative potential and limited effects on DNA methylation. Culturing BMSCs in xenogenic serum resulted in loss of function during cartilage disk formation and increased clotting risk.

The results show that LVAD treatment of decompensated end-stage PPCM is feasible, and early LVAD provision can lead to safe LV recovery in young female patients.

Chondrocyte Isolation from Loose Bodies-An Option for Reducing Donor Site Morbidity for Autologous Chondrocyte Implantation

Summary: This study found that loose bodies (LBs) from patients with osteochondritis dissecans (OCD) contain sufficient viable and functional chondrocytes that can be used for autologous chondrocyte implantation (ACI) without affecting their phenotype during prolonged in vitro expansion. The chondrocyte populations from LBs (LB-CH) showed no significant differences in cell yield per isolation and expression of chondrocyte progenitor cell markers compared to control chondrocytes from non-weight-bearing joint regions. LB-CH exhibited comparable viability, proliferation rates, and chondrogenic differentiation potential to control chondrocytes, and had a higher ability to form a hyaline cartilage matrix compared to bone marrow mesenchymal stromal cells (BMSCs).

The Telomerase Connection of the Brain and Its Implications for Neurodegenerative Diseases

Summary: Telomerase, consisting of TERT and TERC, is well-known for maintaining and extending telomeres. However, TERT has non-canonical functions, including shuttling to the cytoplasm and mitochondria to protect cells from stress. TERT also plays a protective role in brain neurons by countering effects of toxic proteins and activating protein degradation pathways, suggesting its potential in treating neurodegenerative diseases.

A Novel Fluorescence-Based Screen of Gene Editing Molecules for Junctional Epidermolysis Bullosa

Summary: In this study, a cell line suitable for gene expression studies of the JEB-associated COL17A1 gene was developed using the CRISPR/Cas9 system. The accurate expression and localization of the GFP-C17 fusion protein were confirmed. Repairing a JEB-associated frameshift mutation restored the expression and localization of the fusion protein. This fluorescence-based JEB cell line has the potential to be used for personalized gene editing and applications in vitro and in animal models in vivo.
