Summary: This study evaluates the impact of protocatechuic acid (PCA) on glucose and lipid dysregulation induced by a high-fat diet. The results show that PCA treatment improves systemic insulin resistance, reduces ectopic lipid accumulation, and enhances insulin action in the liver and peripheral tissues.

Summary: The tumor suppressor p53 plays a crucial role in controlling liver glycogen storage and is linked to insulin signaling through the transcription factor FOXO1. Acute deletion of p53 prevents glycogen accumulation upon refeeding, while chronic loss of p53 leads to compensational activation of glycogen synthesis pathway. Additionally, high-fat diet-induced insulin resistance with persistent FOXO1 activation affects the regulation of p53 and its target genes during fasting, showing overlapping effects of both FOXO1 and p53 on gene expression in a context-dependent manner.

Summary: Obesity is a global public health concern that hinders socio-economic development and human productivity. Polyphenols have emerged as a promising solution in combating obesity due to their lower side effects compared to synthetic pharmaceuticals. This study reviews the potential anti-obesity effects of dietary polyphenols, focusing on their biological activities, influence on energy metabolism and mechanisms. It highlights their roles in various processes such as glucose uptake, lipid metabolism, and mitochondrial regulation. The findings confirm that polyphenols can prevent and ameliorate obesity through multiple mechanisms, including fighting insulin resistance, scavenging reactive oxygen species, and regulating key enzymes.

Summary: Early childhood malnutrition may lead to the onset of obesity and diabetes mellitus in adulthood. The study found that tauroursodeoxycholic acid (TUDCA) has beneficial effects on glucose homeostasis and body fat accumulation in protein-restricted mice. TUDCA reduces obesity, improves metabolic flexibility, and restores glucose tolerance and insulin sensitivity through multiple pathways, serving as a potential strategy to reverse metabolic disorders.

Summary: Lipid and glucose metabolism are crucial for human activities and their dysregulation can lead to diabetes and obesity. Recent studies have shown that the bone, acting as an endocrine organ, plays a role in systemic metabolism through bone-derived molecules. Sclerostin, a protein produced by osteocytes, not only inhibits bone formation but also affects lipid and glucose metabolism. This paper provides a comprehensive review of the effects of sclerostin on lipid and glucose metabolism, discusses the progress in sclerostin research, and explores its potential manipulation for obesity and diabetes treatment.

Summary: Peripheral dopamine plays a role in regulating glucose uptake in insulin-sensitive tissues, with dopamine receptors having differential involvement. Dopamine also affects lipid metabolism in white adipose tissue. The modulation of the dopaminergic system in the periphery could be a potential therapeutic approach for metabolic disorders.

Summary: By selectively knocking out HIF1a in the neurons of the mediobasal hypothalamus, mice showed significant increases in body weight and food intake, as well as abnormalities in glucose and lipid metabolism. Insulin-induced AKT phosphorylation levels decreased and Glut4 mRNA expression was reduced in the absence of hypothalamic neuronal HIF1a.

Summary: The study found that gentiopicroside (GPS) can activate the PI3K/AKT pathway by promoting DNA-binding protein 2 (DDB2)-mediated degradation of progestin and adipoQ receptor 3 (PAQR3), thereby improving glycolipid metabolism. GPS directly binds to PAQR3 and inhibits the interaction between PAQR3 and the PI3K catalytic subunit (P110a) to restore insulin signaling pathway.

Summary: The transcription factor FoxO1 regulates adipose transdifferentiation by mediating Tgf beta 1 signaling. Loss of FoxO1 induces browning of adipose tissue and increases energy expenditure. The FoxO1-Tgf beta 1 signaling cascade also plays a role in adipose browning induced by beta 3-AR agonist.
