Blood cells transcriptomics as source of potential biomarkers of articular health improvement: effects of oral intake of a rooster combs extract rich in hyaluronic acid

The aim of the study was to explore peripheral blood gene expression as a source of biomarkers of joint health improvement related to glycosaminoglycan (GAG) intake in humans. Healthy individuals with joint discomfort were enrolled in a randomized, double-blind, placebo-controlled intervention study in humans. Subjects ate control yoghurt or yoghurt supplemented with a recently authorized novel food in Europe containing hyaluronic acid (65 %) from rooster comb (Mobilee (TM) as commercial name) for 90 days. Effects on functional quality-of-life parameters related to joint health were assessed. Whole-genome microarray analysis of peripheral blood samples from a subset of 20 subjects (10 placebo and 10 supplemented) collected pre- and post-intervention was performed. Mobilee (TM) supplementation reduced articular pain intensity and synovial effusion and improved knee muscular strength indicators as compared to placebo. About 157 coding genes were differentially expressed in blood cells between supplemented and placebo groups post-intervention, but not pre-intervention (p < 0.05; fold change >= 1.2). Among them, a reduced gene expression of glucuronidase-beta (GUSB), matrix metallopeptidase 23B (MMP23B), xylosyltransferase II (XYLT2), and heparan sulfate 6-O-sulfotransferase 1 (HS6ST1) was found in the supplemented group. Correlation analysis indicated a direct relationship between blood cell gene expression of MMP23B, involved in the breakdown of the extracellular matrix, and pain intensity, and an inverse relationship between blood cell gene expression of HS6ST1, responsible for 6-O-sulfation of heparan sulfate, and indicators of knee muscular strength. Expression levels of specific genes in blood cells, in particular genes related to GAG metabolism and extracellular matrix dynamics, are potential biomarkers of beneficial effects on articular health.

Breast Milk MicroRNAs Related to Leptin and Adiponectin Function Can Be Modulated by Maternal Diet and Influence Offspring Phenotype in Rats

Summary: This study investigated the effects of maternal diet during lactation on the expression of leptin, adiponectin, and related miRNAs in breast milk, as well as their impact on dams and their offspring. The results showed that a diet enriched with oleic acid, betaine, and leucine during lactation can affect the ratio of leptin to adiponectin in breast milk, leading to increased body fat in offspring and higher insulin resistance in adulthood. Additionally, specific miRNAs in breast milk were also influenced by the maternal diet.

miR-222 exerts negative regulation on insulin signaling pathway in 3T3-L1 adipocytes

Summary: Elevated miR-222 levels are linked to metabolic syndrome, insulin resistance, and diabetes. Rats fed an obesogenic diet during lactation have higher miR-222 content in breast milk, leading to increased body fat mass and impaired insulin sensitivity in their offspring. Transfection experiments with miR-222 mimics or inhibitors on 3T3-L1 preadipocytes reveal that miR-222 plays a role in pathways related to insulin signaling, lipid metabolism, and adipogenesis.

Influence of Maternal Metabolic Status and Diet during the Perinatal Period on the Metabolic Programming by Leptin Ingested during the Suckling Period in Rats

Summary: The study aimed to analyze the long-term metabolic effects of leptin supplementation during suckling in the offspring of diet-induced obese rats and the potential benefits of improving maternal diet during lactation. The results showed that leptin treatment significantly improved metabolic profile and body fat accumulation under standard diet conditions, with stronger effects in male offspring. However, under high-fat diet conditions, the offspring of dams fed as high-fat diet showed metabolic alterations that were not observed in the offspring of dams switched to standard diet during lactation. Leptin supplementation improved glucose homeostasis in male offspring but impaired insulin sensitivity in female offspring born to high-fat diet-fed dams. These findings highlight the importance of balanced maternal diet during the perinatal period, especially lactation, for the metabolic health of offspring, and the beneficial effects of leptin supplementation during suckling, particularly in male offspring.

Summary: The study evaluated the impact of improving maternal diet during lactation on the metabolome of milk and offspring plasma in diet-induced obese rats. The results showed that the milk of rats fed a western diet had altered amino acid and carnitine profiles, as well as changes in other metabolites. These alterations were also observed in the plasma of the offspring, but were largely normalized in rats that switched to a standard diet during lactation.
