Summary: This study aims to investigate the mechanism of MCL-1i-induced Mcl-1 protein stability. Both in vivo and in vitro experiments demonstrate that MCL-1i-induced Mcl-1 protein stability is mainly associated with defective Mcl-1 ubiquitination and concurrent apoptosis induction. Additionally, MCL-1i also enhances Mcl-1Thr163 phosphorylation via ERK pathway, contributing to Mcl-1 stability.

Summary: The engineering of oncolytic vaccinia virus (VV) with immune checkpoint blockade, such as VV-scFv-TIGIT, demonstrated enhanced antitumor efficacy by reshaping the suppressive tumor microenvironment and synergizing with PD-1 or LAG-3 blockade to achieve complete response in tumors with poor response to VV or immune checkpoint blockade monotherapy.

Summary: The E3 ubiquitin ligase TRIP12 is required for efficient mitosis and completion of cell division in cancer cells. Inhibition of TRIP12 enhances Taxol-induced cell death, depending on the action of FBW7 and MCL-1. Therefore, the TRIP12/FBW7/MCL-1 axis plays a crucial role in the response of cancer cells to Taxol.

Summary: Intracellular Ca2+ signals play a crucial role in various cellular processes. Research has shown that BAPTAi can induce apoptosis in cancer cells by inhibiting mTORC1 activity and impairing glycolysis, independent of Ca2+ signaling. Additionally, direct inhibition of PFKFB3 emerges as a potential therapeutic target in MCL-1-dependent cancers.

Summary: BCL-2, the first oncogene described to promote cell survival, continues to be a popular therapeutic agent in oncology as the prime target of venetoclax. However, in hematology, its pro-survival sibling MCL-1 is now taking the spotlight, as elegantly demonstrated in the latest issue of Blood.

Summary: Leukemia or lymphoma cells with TP53 loss have a selective advantage when treated with sublethal doses of a BCL-2 or MCL-1 inhibitor due to reduced activation of BAX and BAK, but this defect can be overcome with dual treatment with both inhibitors.

Summary: The authors developed a mathematical model based on fractional order ordinary differential equations to investigate how virotherapy could enhance chemotherapy under the action of MEK inhibitor. The results showed that virus burst size and MEK inhibitors have a noticeable impact on regulating the trend of tumor cell proliferation. This study's findings can contribute to the development of a therapeutic strategy that combines MEK inhibitor functional monitoring with tumor cell growth control.
