Summary: In hepatocellular carcinoma (HCC), PD-L1(+) host macrophages are a major cellular source of PD-L1 in tumors, displaying a glycolytic phenotype and producing antitumor IL-12p70. The regulation of these macrophages involves fibronectin 1-induced PKM2, controlling both antitumor properties and inflammation-mediated PD-L1 expression. Modulating the context of glycolytic macrophages in HCC tumors may restore their antitumor properties and offer a precise strategy for anticancer therapy.

Summary: A cascade-amplified PTT/CDT nanoplatform was constructed by coating IR780-embedded nanoparticles and glucose oxidase-loaded Fe3O4 nanoparticles on red blood cell membranes for effective HCC treatment. The nanoplatform interfered with glucose metabolism to sensitize the PTT and accelerated the Fenton reaction for enhanced CDT, providing an alternative strategy for effective tumor treatment.

Summary: This study investigates the prognostic value of transcriptomes associated with tumor FDG uptake in hepatocellular carcinoma (HCC). Through analyzing the correlations between metabolic genes and FDG uptake, multiple genes related to glycolysis and fatty acid metabolism were identified. A metabolic gene expression balance score was established and showed good prognostic value in HCC. These findings highlight the potential use of FDG PET/CT imaging biomarkers in selecting appropriate patients for metabolism-targeted therapy in HCC.

Summary: In this study, the dysregulation of fatty acid and glucose metabolic pathways in liver tumors was investigated. The results showed that fatty acid metabolism was highly expressed in early stages of liver tumors, while glucose metabolism was prominently activated in hepatocellular carcinoma (HCC) stage. The dysregulation of these metabolic pathways was further confirmed in recurrent cirrhosis and recurrent HCC samples. The expression of genes related to fatty acid and glucose metabolism had significant predictive value for overall survival and recurrence-free survival in HCC patients.
