Summary: This study found that alpha-PVP, a novel psychoactive substance, impairs spatial learning/memory and brain mitochondrial function in rats at high doses. The research observed a decline in spatial learning/memory and dysfunction of brain mitochondria after 10 days of repeated administration of different doses of alpha-PVP. These findings provide the first evidence of impaired spatial learning/memory and the potential role of brain mitochondrial dysfunction in cognitive impairments caused by repeated use of alpha-PVP.

Summary: Mass spectrometry imaging (MSI) is a powerful technique that can reveal the spatial distribution of atomic and molecular ions on the surfaces of biological samples. MALDI-TOF and TOF-SIMS, two commonly used MSI techniques, have witnessed significant advancements in studying cells and tissue sections due to their high resolution, sensitivity, precision, and chemical specificity. This article briefly discusses the principles and sampling methods of MALDI-TOF and TOF-SIMS, while highlighting their importance as diagnostic tools and robust analytical techniques in the medicinal, pharmaceutical, and toxicology fields through representative published studies.

Summary: The study evaluated the mutagenicity of Mexedrone, alpha-PVP and alpha-PHP on DNA, indicating Mexedrone's mutagenic potential while the other two compounds did not show mutagenicity; even with extrinsic metabolic activation, the mutagenic capacity of alpha-PVP and alpha-PHP could not be excluded, and their metabolites were found to be mutagenic. The induction of reactive oxygen species (ROS) was evaluated as a possible mechanism for the effects but did not show a statistically significant increase in ROS levels for any of the tested substances.

Summary: Increasing reports of neurological and psychiatric outcomes due to the use of psychostimulant synthetic cathinones (SCs) have raised public concern. However, the neurotoxic mechanisms of a major MDPV derivative, alpha PHP, remain poorly understood. In this in vitro study, alpha PHP was found to significantly decrease the viability, proliferation, and clonal capability of murine neural stem/progenitor cells, accompanied by activation of apoptotic/autophagic/necroptotic pathways and ultrastructural alterations. These findings suggest that alpha PHP may damage neural stem/progenitor cells and the fundamental units of adult neurogenic niches, leading to long-lasting and irreversible CNS damage. This research is important for gaining a broader understanding of the toxicology of SCs and establishing appropriate treatment for new psychoactive substances (NPS) and their potential consequences for public health.
