Cultivation of circulating tumor cells in esophageal cancer

The presence of circulating tumor cells (CTCs) in patients with metastatic carcinoma is generally associated with poor clinical outcome. There have been many investigations showing a possible use of CTCs as minimally invasive predictive and prognostic biomarker in cancer medicine. In this report a size-based method (MetaCell (R)) for quick and easy enrichment and cultivation of CTCs is presented to enable possible CTCs use in esophageal cancer (EC) management. In total, 43 patients with diagnosed EC, 20 with adenocarcinoma (AdenoCa) and 23 with squamous cell carcinoma (SCC), were enrolled into the adaptive prospective-like study. All the patients were candidates for surgery. The CTCs were detected in 27 patients (62.8%), with a higher rate in adenocarcinoma (75%) than SCC (52%). Finally, there were 26 patients with resectable tumors exhibiting CTCs-positivity in 69.2% and 17 patients with non-resectable tumors with 41.7% CTCs-positivity. Interestingly, in the patients undergoing neoadjuvant therapy, the CTCs were detected at time of surgery in 55.5% (10/18). The overall size-based filtration approach enabled to isolate viable CTCs and evaluate to their cytomorphological features by means of vital fluorescent staining. The CTCs were cultured in vitro for further downstream applications including immunohistochemical analysis. This is the first report of the successful culturing of esophageal cancer CTCs. The detection of CTCs presence could help in the future to guide timing of surgical treatment in EC patients.

Summary: This paper presents a case of a 36-year-old patient initially diagnosed with extensive endometriosis involving the anterior abdominal wall and the pelvis minor. After biopsy, a diagnosis of advanced low-grade serous ovarian cancer was established. Novel Treatment Options in Metastatic Esophageal Carcinoma: Checkpoint Inhibitors in Combination Therapies

Summary: A systematic review was conducted to evaluate the potential of immune checkpoint inhibitors (ICIs) combined with chemotherapy, radiotherapy, and/or biologic therapy in metastatic esophageal carcinoma (EC) patients. The review discussed the effects of different treatment approaches on the tumor microenvironment and reviewed the current clinical trials to provide a rationale for immunotherapy application in combination settings. PD-L1 expression was found to be higher in squamous cell carcinoma (SCC) compared to adenocarcinoma, suggesting that ICIs plus chemotherapy may be a feasible option for SCC patients. Radiation was shown to induce immune checkpoint molecules and activate cytotoxic T cells, providing a rationale for ICI administration in combination with radiotherapy. The combination of ICIs with biologic therapy was deemed safe, but its impact on clinical outcomes varied.

Summary: This study aimed to evaluate the effectiveness of VR therapy on improving the mental state, quality of sleep, and physical activity of breast cancer patients. The results showed that VR therapy can significantly reduce the severity of depressive symptoms, decrease destructive coping styles, and have a positive impact on constructive coping styles.

Prolactin-induced protein (PIP) increases the sensitivity of breast cancer cells to drug-induced apoptosis

Summary: This study demonstrates that high expression of prolactin-induced protein (PIP) sensitizes breast cancer (BC) cells to anti-cancer drugs. Increased sensitivity to chemotherapy is the result of pro-apoptotic activity of PIP, which is evidenced by up-regulation of specific pro-apoptotic genes. The findings suggest that PIP can serve as a marker for the prognostic evaluation of adjuvant chemotherapy.

Effect of targeted coagulopathy management and 5% albumin as volume replacement therapy during lung transplantation on allograft function: a secondary analysis of a randomized clinical trial

Summary: After lung transplantation, primary graft dysfunction (PGD) contributes to early postoperative morbidity. Intraoperative transfusion of blood products and ischemia-reperfusion injury play a role in PGD development. A randomized clinical trial showed that targeted coagulopathy management and intraoperative administration of 5% albumin can reduce blood loss during lung transplantation surgery. The analysis further revealed that the POC group had better graft function and lower dosage of norepinephrine compared to the non-POC group. The difference in 1-year survival was not significant.

Summary: This study investigates the differential effects of breast cancer subtypes on T cell mediated immune responses through the isolation of EVs from patient serum and cell-line derived EVs. The results show that TNBC patients have the lowest number of EVs in their serum, while ER+HER2+ patients have the highest. Furthermore, EVs derived from different breast cancer subtypes have different immunomodulatory capabilities, with TNBC EVs demonstrating the most immune-suppressive phenotype.

The impact of cellular elements of TME on melanoma biology and its sensitivity to EGFR and MET targeted therapy

Summary: The microenvironment of melanoma, consisting of cellular elements and physicochemical conditions, plays a significant role in melanoma progression, proliferation, metabolism, and response to anti-cancer therapy. Fibroblasts, adipocytes, and keratinocytes present in the tumor niche influenced melanoma cells' signaling pathways, metabolism, and proliferation. The findings indicate the efficacy of the proposed combination therapy for melanoma remains even when cancer-associated cells are present in the vicinity.

Summary: This study aimed to investigate the effects of Oncostatin M (OSM) on immune cells derived from liposarcoma, leiomyosarcoma, and myxofibrosarcoma, and the potential cooperation between OSM and nivolumab in treating these soft tissue sarcomas (STSs). The results suggest that OSM may have a role in the treatment of leiomyosarcoma, myxofibrosarcoma, and liposarcoma. However, further histotype-tailored studies are needed to fully comprehend the functions of OSM in STSs.
