Four new dimeric sesquiterpene lactones (japonicones Q-T, 1-4) were isolated from the aerial part of Inula japonica Thunb., together with two known ones. Their structures were determined mainly by 1D and 2D NMR spectroscopic techniques including HSQC, H-1-H COSY, HMBC, and NOESY. Herein, the inhibitory activities against lipopolysaccharide (LPS)-induced nitric oxide (NO) production in RAW264.7 macrophages of 1-6 were evaluated; IC50 values were tested as 8.5, 8.9, 4.3, 4.3, 4.2, and 9.2 mu M, respectively. Summary: A novel cyclic tripeptide, sclerotiotide M (1), was isolated from the culture of marine derived fungus Aspergillus ochraceopetaliformis DSW-2, along with four known compounds. The structure of sclerotiotide M (1) was determined through various analytical methods, and it, along with the other compounds, exhibited weak cytotoxic activities against cancer cell lines HPAC and BXPC3.

Predicting hERG channel blockers with directed message passing neural networks

Summary: In this study, a classification model for identifying hERG blockers was developed using the D-MPNN model based on diverse datasets, and the D-MPNN + moe206 model showed significantly improved performance. Our results indicated that the D-MPNN + moe206 model is among the best classification models and has potential applications in the discovery of novel and effective hERG blockers.

Drug-resistant HER2-positive breast cancer: Molecular mechanisms and overcoming strategies

However, resistance to targeted therapy is an issue that needs to be addressed.

Glutathione peroxidase 2 knockdown suppresses gastric cancer progression and metastasis via regulation of kynurenine metabolism

Summary: Gastric cancer (GC) is a deadly malignancy that requires new therapeutic targets for effective treatment. Glutathione peroxidase-2 (GPx2) was found to be overexpressed in clinical GC samples and negatively correlated with prognosis. Knockdown of GPx2 inhibited GC progression and metastasis by suppressing the KYNU-kyn-AhR signaling pathway, which was activated by reactive oxygen species (ROS) accumulation.

Their structures were determined using spectroscopic analysis, single crystal X-ray diffraction, quantum chemical 13C NMR, and electronic circular dichroism calculations. All compounds exhibited potent inhibitory activities against indoleamine 2,3-dioxygenase and tryptophan-2,3-dioxygenase in enzymatic assays, with compound 2 showing the highest activity.

Summary: The relevance of oxylipins as biomarkers of oxidative stress has been established in recent years. This study evaluated the lipidomic profiling of different aerial parts of Acacia cyanophylla Lindl. using LC-MS/MS technology for the first time. The analysis revealed the existence of six PhytoPs and three PhytoFs, with stems having the highest amount of these metabolites. This study provides a foundation for exploring Acacia cyanophylla Lindl. as a potential source of plant oxylipins for therapeutic or pharmaceutical uses.
