Calcineurin (CN), a multifunctional protein, mediates the immune response through diverse signaling pathways in mammals, while the function of CN in the immune response of molluscan hemocytes still remains unclear. In the present study, we detected the distribution of CN in various tissues and the expression levels of Pf-CNA and Pf-CNB gene in hemocytes of Pinctada fucata. After the preparation of hemocyte monolayers, we checked the response of enzymatic activity of CN, the degradation level of I kappa B alpha, the activity of iNOS and the production of NO, and IL-2 to the challenge of lipopolysaccharide (LPS) and cyclosporin A (CsA). CN activity in hemocytes was very sensitive to both the stimulation of LIPS and the inhibition of CsA. Most importantly, I kappa B alpha degradation in hemocytes was induced by LPS and attenuated by CsA. Consequently, the activity of iNOS was elevated and the production of NO was increased. Additionally, we found that the synthesis of IL-2 was increased by LPS but was apparently weakened by CsA. In vivo bacterial clearance experiments showed that CsA significantly decreased the ability of in vivo bacteria clearance in pearl oyster. All the results revealed, for the first time, that CN mediated the immune response of molluscan hemocytes via activating NF-kappa B signaling pathway. (C) 2009 Elsevier Ltd. All rights reserved.

A novel matrix protein PNU5 facilitates the transformation from amorphous calcium carbonate to calcite and aragonite

Bowen Shuai, Tingyue Deng, Liping Xie, Rongqing Zhang

Summary: In this study, a novel matrix protein named PNU5 was discovered to play a regulatory role in both prismatic layer and nacreous layer formation in Pinctada fucata. Functional studies showed that PNU5 might be involved in shell formation in a positive manner. Repression of pnu5 affected the structure of both layers, while the recombinant protein rPNU5 enhanced the precipitation rate of CaCO3 and altered the morphology of the crystals.

A novel bifunctional protein PNU7 in CaCO3 polymorph formation: Vaterite stabilization and surface energy minimization

Li Yi, Bing Zou, Liping Xie, Rongqing Zhang

Summary: This article investigates a novel vaterite stabilizer acidic matrix protein PNU7, which is found in both the prism and nacre of Pinctada fucata. The study shows that PNU7 can trigger the formation of stable vaterite even with a deficiency in Mg2+ and low Ca2+ levels. Moreover, increasing PNU7 in the calcium carbonate crystallization system leads to a decrease in crystal size and an increase in nucleation quantity. The results suggest that PNU7 not only plays a role in shell formation but also regulates the formation of nacre through surface energy minimization.

Elemental and boron isotopic variations in tourmaline in two-mica granite from the Cuona area, Tibet: Insights into the evolution of leucogranitic melt

Guozhi Xie, Haibo Yan, Guangming Li, Jia Guo, Fang Liu, Qian Chen, Rongqing Zhang, Lipeng Zhang

Summary: This study focused on the in-situ major and trace element and boron isotope compositions of tourmalines from two-mica granites in the Cuona area. The results indicated that tourmalines in the Cuonadong leucogranite crystallized earlier than those in the Cuona leucogranite during magmatic differentiation. The volatile contents of tourmaline were genetically related to incompatible elements, which led to tourmaline zonation and the enrichment of Sr, Zn, and Pb. The B isotopic composition of tourmaline suggested that the granitic magma was dominantly derived from the partial melting of the metasedimentary source rocks, and their B isotope variations were likely due to fluid exsolution during B-rich melt evolution. High rare metal contents in tourmalines indicated great mineralization potential in the two-mica granites of the Cuona area.

Extraction of bioactive compounds from cinnamon residues with deep eutectic solvents and its molecular mechanism

Rongqing Zhang, Haifeng Chen, Qiang Yu, Yu Zhang, Fen Liu, Fan Wang, Xiaoyan Chen, Yunyun Liu

Summary: The recycling of bioactive compounds from Chinese herb residue is hindered by low extraction efficiency and high energy environmental impact. A microwave-assisted deep eutectic solvent (DES) extraction method was developed to enhance the extraction yield of bioactive compounds from cinnamon twigs residue. The optimal DES system of choline chloride/ethylene glycol achieved a significant increase in extraction yield, with a 41.528% and 41.443% improvement for cinnamic acid and cinnamic aldehyde, respectively, compared to traditional extraction processes. The extraction process was found to be a spontaneous, exothermic, and irreversible reaction, following a second-order kinetic model. Quantum chemistry analysis revealed that the extraction capacity relied mainly on hydrogen bonding force, and the difference in binding energy between the solvent and the target compounds explained the variation in extraction yields.

Direct control of shell regeneration by the mantle tissue in the pearl oyster Pinctada fucata

Jingliang Huang, Yangjia Liu, Taifeng Jiang, Wentao Dong, Liping Xie, Rongqing Zhang

Summary: Molluscs rapidly repair damaged shells to ensure survival, but the control mechanism of this process is unclear. Using scanning electron microscope and histochemical analysis, this study examined shell regeneration in Pinctada fucata. Damage to the shell caused the mantle tissue to retract, resulting in the rearrangement of mantle zones and the alteration of shell morphology. The up-regulation of shell matrix protein genes and the change in CaCO3 dynamics promoted shell formation. This study reveals the relationship between mantle tissue activities and regenerated shell morphology.

Lingyun Zhou, Wenqiang Pu, Ming-yi You, Rongqing Zhang, Qingjiang Shi

Summary: In this paper, the problem of cooperative sensing via a system of multi-unmanned aerial vehicles (UAVs) is considered. Each UAV is equipped with a directional antenna to perform detection tasks for multiple targets. The detection probability is chosen as the metric to measure the system's perception ability, and the goal is to maximize the sum detection probability by optimizing UAVs' deployment and directional antenna orientations. To tackle the nonconvexity of the problem, it is decomposed into two sub-problems and efficient algorithms are developed using block coordinate descent and Gibbs Sampling approaches.
