Chronic exposure to the parasite Enteromyxum leei (Myxozoa: Myxosporea) modulates the immune response and the expression of growth, redox and immune relevant genes in gilthead sea bream, Sparus aurata L.

The myxosporean parasite Enteromyxum leei invades the intestine of gilthead sea bream producing a slow-progressing disease, which may end in the death of fish. The present work aimed to better know the host immune response and the underlying molecular mechanisms, which may help to understand why some individuals seem to be refractory to the disease. Three main aspects involved in fish health and welfare (immune, growth and redox status) were studied in fish exposed to E. leei-contaminated effluent, in comparison with control animals (not exposed to the disease). After chronic exposure (113 days), prevalence of infection was 67.8%. Among exposed fish, parasitized and non-parasitized fish exhibited clear differences in some of the measured innate immune factors (respiratory burst, serum peroxidases, lysozyme and complement), and in the expression of immune, antioxidant and GH-related genes. The respiratory burst of parasitized fish was significantly higher, and serum peroxidases and lysozyme were significantly decreased both in parasitized and non-parasitized fish. The gene expression of GHR-I, GHR-II, IGF-I and IGF-II was measured in head kidney (HK) samples, and that of interleukin (IL)-1 beta, tumour necrosis factor (TNF)-alpha, alpha-2M, GR, GPx-1 and GRP-75 was measured in intestine and HK samples, by rtqPCR. Parasitized fish exhibited a down-regutation of IL-1 beta, TNF-alpha and GPx-I in the intestine, and GHR-I and IGF-I were also down regulated in HK. alpha-2M and GRP-75 were over-expressed in the intestine of parasitized animals. Non-parasitized fish had increased transcripts of GHR-I and IGF-I with respect to control animals, which could furnish their immunocytes with an advantage to combat the parasite. Summary: The GPRO suite is an ongoing bioinformatic project for the analysis of -omics data. It introduces a client- and server-side solution for comparative transcriptomics and variant analysis. The client-side includes two Java applications, RNASeq and VariantSeq, which manage pipelines and workflows using common command line tools. The server-side, named GPRO Server-Side, hosts all dependencies and requires a Linux operating system, PHP, SQL, Python, bash scripting, and third-party software. The solution offers both desktop and web applications, with step-by-step and pipeline modes for execution. It also features an experimental support system called GENIE, with a chatbot assistant and expert system for troubleshooting and recommendations.

Fish microbiomics: Strengths and limitations of MinION sequencing of gilthead sea bream (Sparus aurata) intestinal microbiota

Summary: This study compared different sequencing platforms for microbial characterization in fish gut mucosal samples. The results showed that MinION is a reliable and accurate tool for assessing intestinal bacteria communities, with good correlation with Illumina results. However, less abundant but biologically important taxa may be masked compared to Illumina sequencing.

A New Parasitic Archamoeba Causing Systemic Granulomatous Disease in Goldfish Extends the Diversity of Pathogenic Endolimax spp.

Summary: A new parasite species, Endolimax carassius, was identified as the causative agent of systemic granulomatous disease in goldfish. Phylogenetic analyses showed that this goldfish parasite species is closely related to Endolimax piscium, the causative agent of systemic granulomatous disease in Senegalese sole. The discovery of a considerable diversity of Endolimax spp. among fish highlights the importance of characterizing these parasites in order to understand their evolution and pathogenic potential.

Sparicotyle chrysophrii experimental infection of gilthead seabream (Sparus aurata): Establishment of an in vivo model reproducing the pathological outcomes of sparicotylosis

Summary: This study explores the best experimental conditions to set up an in vivo infection model capable of mimicking the sparicotylosis signs observed in farmed diseased fish. The experimental setup for parasite transmission consisted of a recipient fish tank with naive GSB receiving water from two S. chrysophrii-infected donor tanks. The study provides an advancement for studying S. chrysophrii's biology and its interaction with its host, as well as for further researching treatment alternatives and prophylactic measures.

Summary: This study combines multiple systems to understand the schooling behavior and adaptive features of farmed gilthead sea bream in high stocking density conditions. The results show that high density fish exhibit adaptive behavior, such as changes in feeding behavior and enhanced endogenous swimming activity rhythm, but also experience reduced growth rates and limited oxygen availability.

Summary: This paper introduces SAMBA, a computational tool that uses a Bayesian network approach to model the network structure and interactions of fish gut microbiomes in aquaculture systems. SAMBA can infer the influence of farming conditions on microbiome diversity and predict changes under different conditions. It is worth noting that SAMBA can be used for modeling microbiome-host network relationships of any vertebrate organism, including humans, in any system and/or ecosystem.
