Summary: In this study, a new therapeutic target for the treatment of leishmaniasis was discovered using an activity-based protein profiling strategy to investigate serine proteases (SPs) in Leishmania parasites. Significant differences in the expression levels of active Leishmania serine hydrolases (SHs) were observed throughout the parasite's life cycle and between different species. Two targetable SPs were identified in Leishmania mexicana using quantitative proteomic mass spectrometry. The results of this study provide templates for the discovery of new drugs for the treatment of leishmaniasis.

Summary: This study found that Bacillus licheniformis TG116 isolated from Typhonium giganteum Engl. can produce a stable fermentation supernatant that inhibits the growth of several plant pathogens. The fermentation supernatant has protease and cellulase activities and is stable at various temperatures and under UV exposure. The antifungal protease produced by TG116 was purified and its inhibition by surfactants was observed. The presence of a serine protease gene was confirmed in strain TG116.

Summary: The Kunitz-Soybean Trypsin Inhibitor (Kunitz-STI) family is a large family of proteins with most of its members being protease inhibitors. This study analyzed the crystallographic structures of Kunitz-STI inhibitors to characterize the structural features and locations of serine protease-binding loops. The findings suggest a relationship between the conformation of these loops, the inhibition mechanism, and the plant source of the inhibitors. This research challenges the perception that all Kunitz-STI inhibitors are canonical and will aid in the identification and design of multifunctional proteins.

Summary: Elafin and its precursor trappin-2 play an important role in protecting the intestinal mucosal barrier from microbial invasion, especially in gut inflammation. They have shown protective effects in gut intestinal disorders associated with acute or chronic inflammation, as well as gluten sensitization disorders, through various mechanisms. The potential therapeutic application of delivering trappin-2/elafin to the intestinal mucosa surface and its role in intestinal tissue repair are also discussed.

Summary: This study evaluated the morphological changes caused by fractions and subfractions obtained from barks of Aspidosperna nitidum against L. (L.) amazonensis promastigotes. The results suggest that alkaloids isolated from A. nitidum show potential as leishmanicidal agents. Scanning electron microscopy and transmission electron microscopy analysis revealed changes in the parasites' morphology after treatment with the active fractions.

Summary: The study designed and synthesized 32 new M-pro inhibitors containing bicycloproline, which showed inhibitory effects on SARS-CoV-2. Compounds MI-09 and MI-30 exhibited excellent antiviral activity in cell-based assays and significantly reduced lung viral loads and lung lesions in a transgenic mouse model of SARS-CoV-2 infection. Both also displayed good pharmacokinetic properties and safety in rats.
