Summary: The expression of SLPI is significantly down-regulated in HCC tissues and cell lines. Low level of SLPI predicts worse survival in HCC. Silencing of SLPI promotes HCC cell proliferation, migration and invasion, while overexpression of SLPI suppresses these abilities. SLPI plays a protective role in HCC progression by activating ER stress-mediated apoptosis of hepatoma cells through MAPK signaling pathways.

Summary: Elafin and its precursor trappin-2 play an important role in protecting the intestinal mucosal barrier from microbial invasion, especially in gut inflammation. They have shown protective effects in gut intestinal disorders associated with acute or chronic inflammation, as well as gluten sensitization disorders, through various mechanisms. The potential therapeutic application of delivering trappin-2/elafin to the intestinal mucosa surface and its role in intestinal tissue repair are also discussed.

Summary: This study demonstrates that excessive dietary iron intake perturbs diet-microbiota-epithelium interactions, leading to the initiation of intestinal tumors. The gut microbiota modulated by excessive dietary iron promotes colorectal tumorigenesis by increasing the permeability of the gut barrier and causing leakage of lumen bacteria. Epithelial cells respond to the leaked bacteria by releasing more secretory leukocyte protease inhibitor (SLPI), which acts as a pro-tumorigenic factor by activating the MAPK signaling pathway. Furthermore, the depletion of Akkermansiaceae in the gut microbiota due to excessive dietary iron can be attenuated by supplementation with Akkermansia muciniphila.

Summary: The study identified SLPI as a highly upregulated cellular protease inhibitor in chondrocytes treated with pathogenic catabolic factors associated with osteoarthritis (OA). Although SLPI does not directly modulate OA development in mice, it may serve as a potential biomarker of OA in humans and animal models, as its levels were significantly elevated in human OA patients and experimental OA mice.

The results showed that increased concentrations of SLPI in both blood serum and endometrium were associated with poor prognosis. Summary: This study analyzed the effects of recombinant secretory leukocyte protease inhibitor (rSLPI) on skin wound healing. The results showed that rSLPI can accelerate the wound healing process by increasing CD163 and FGF-2 expression, while decreasing IL-1 and IL-6 expression. This suggests the potential application of rSLPI in improving wound healing and regulating cytokine expression.

Patients with allergy-associated diseases have elevated levels of SLPI in their blood and skin. Exogenous SLPI increases the number of eosinophils and enhances their chemotactic response to CCL11.
