A small-molecule induces apoptosis and suppresses metastasis in pancreatic cancer cells

Pancreatic cancer is one of the most malignant tumor diseases with the characters of aggressive growth and metastasis. With the inefficiency of the current therapeutics, new potential targets and new therapeutic agents for healing of pancreatic cancer are critically needed. We have previously found a small molecule, named 4-tert-butyl-2-[(cyclohexylamino) methyl]-6-methylphenol (TBMMP, NSC number: 48160), which can freeze the intermediate of Ras-GTP hydrolysis in the open non-signaling conformation with high affinity and high specificity in silico. In this work, we studied the effect and mechanism of TBMMP on two pancreatic cancer cell lines, CFPAC-1 and BxPC-3. The results showed that TBMMP could restrain the growth of the pancreatic cancer cells with IC50 value 84.3 mu M for CPFAC-1 and 94.5 mu M for BxPC-3, respectively. Additionally, TBMMP increased cytochrome c release, reduced mitochondrial membrane potential, activated caspase-3, -9, elevated ROS and increased expression of the Bax in the pancreatic cancer cell lines. The results indicated that TBMMP induced the apoptosis of pancreatic cancer cells through the mitochondrial pathway. Further, we also found that TBMMP could suppress the metastasis of both pancreatic cancer cells in vitro. Taken together, we proposed that TBMMP might be a therapeutic potential lead for treating patients with pancreatic cancer. (C) 2013 Elsevier B.V. All rights reserved.

Sensitive and selective detection of Mucin1 in pancreatic cancer using hybridization chain reaction with the assistance of Fe3O4@polydopamine nanocomposites

Summary: Pancreatic cancer is difficult to diagnose in the early stage due to the lack of symptoms. Current diagnosis techniques have limitations in terms of personnel expertise and cost. Therefore, alternative detection methods are highly demanded. In this study, a new strategy combining nanoparticles and amplification reaction was proposed to improve the sensitivity of pancreatic cancer cell detection. The strategy showed good specificity and potential for clinical diagnosis.

Cadmium sulfide as bifunctional mimics of NADH oxidase and cytochrome c reductase takes effect at physiological pH

Summary: Recently, there has been increased attention on the study of mimic enzymes. However, research on the oxidoreductase activity of electron mediators in the biological respiratory chain is still limited. In this study, we found that cadmium sulfide (CdS) nanorods can catalyze the formation of superoxide anions and exhibit NADH oxidase-like activity and Cyt c reductase-like activity. This provides a new perspective for the in-depth study of new nanozymes.

N, P, or S -doped carbon nanotubes as dual mimics of NADH oxidase and cvtochrome c reductase

Summary: In this study, N, P, or S doped carbon nanotubes (CNTs) were used as enzyme mimics of nicotinamide adenine dinucleotide (NADH) oxidase and cytochrome c (Cyt c) reductase. Through N doping, the NADH oxidase-like activity of CNTs was significantly enhanced, with a 4.28-fold increase in the maximum initial velocity for N-doped CNTs (N-CNTs) compared to the unmodified ones. Biologically active NAD* was produced during NADH oxidation, and oxygen was selectively reduced to water or hydrogen peroxide, consistent with natural NADH oxidase. Additionally, it was discovered for the first time that carbon nanotubes can facilitate the electron transfer from NADH to Cyt c, mimicking the properties of Cyt c reductase.

Anwuligan inhibits the progression of non-small cell lung cancer via let-7c-3p/PI3K/AKT/mTOR axis

Summary: Background: Anwuligan (ANW), isolated from nutmeg, has been found to have potential therapeutic effects in diseases. However, its effect on lung cancer is still unclear. Methods: In this study, the cytotoxicity of ANW on non-small cell lung cancer (NSCLC) cells was investigated using various assays. Results: ANW treatment showed clear cytotoxicity against NSCLC cells, inhibiting cell proliferation and migration both in vitro and in vivo. The overexpression of let-7c-3p also inhibited the growth of NSCLC cells. Conclusions: ANW inhibits the growth and metastasis of NSCLC cells by upregulating the expression of let-7c-3p and targeting PIK3CA.

Summary: Most nanozyme research focuses on oxidase and peroxidase capabilities, but this study explores N, P, or S doped carbon nanotubes (CNTs) as enzyme mimics of NADH oxidase and Cyt c reductase. Doping N significantly enhances the NADH oxidase-like activity of CNTs, increasing the maximum initial velocity by 4.28 times. NAD(+) is produced as a biologically active product during NADH oxidation, while oxygen is selectively reduced to water or hydrogen peroxide, similar to natural NADH oxidase. Additionally, carbon nanotubes can promote electron transfer from NADH to Cyt c, mimicking the properties of Cyt c reductase.

Analysis of stabilization mechanisms in β-lactoglobulin-based amorphous solid dispersions by experimental and computational approaches

Summary: Our previous work demonstrated that beta-lactoglobulin-stabilized amorphous solid dispersion (ASD) loaded with 70 % indomethacin remains stable for over 12 months. We further investigated the stabilization mechanisms by testing five other drug molecules and using experimental techniques and molecular dynamics simulations. The results showed that steric confinement, hydrogen bonding, and the glass transition temperature of the drug molecule play important roles in stabilizing ASDs with high drug loadings.

Do the enantiomers of ketamine bind enantioselectively to human serum albumin?

Summary: The binding of drugs to plasma proteins, such as human serum albumin (HSA), is crucial for determining pharmacokinetic parameters. This study investigated the enantioselective binding of S- and R-ketamine to HSA. It was found that ketamine has weak affinity to HSA, with no significant differences in binding behavior between the individual enantiomers and the racemate. The aromatic ring and N-methyl group were identified as the most strongly involved structural moieties in the binding of ketamine to HSA.

Targeting fusion proteins of the interleukin family: A promising new strategy for the treatment of autoinflammatory diseases

Summary: Interleukins (ILs) are important for communication between immune cells and non-immune cells, but dysregulation of ILs expression is a characteristic of autoinflammatory diseases. Drugs targeting ILs have significant clinical benefits, but may also cause adverse reactions. Fusion protein technology, with its ability to enhance therapeutic efficacy, has been explored for developing anti-inflammatory drugs. This review discusses the efficacy of fusion protein drugs developed with ILs or their receptors in treating autoinflammatory diseases, highlighting the potential of this technology in future anti-inflammatory drug development.

The use of polymorphic state modifiers in solid lipid microparticles: The role of structural modifications on drug release performance

Summary: This study investigated the correlation between the structure and release properties of solid lipid microparticles (MPs) with different liquid additives. The additives accelerated the conversion of the unstable alpha-form of tristearin to the stable beta-polymorph and caused structural modifications in the MPs. The presence of additives prolonged the drug release in water and resulted in higher release profiles in biorelevant media. The findings suggest that the release behavior can be influenced by the polymorphism and supramolecular-level structural modification of lipid formulations containing crystal modifiers.

This provides a promising direction for the future development of a safe, effective, and minimally invasive perivascular drug delivery method to treat vascular disease.
