Isobavachalcone [2',4,4'-trihydroxy-3'-(3 ''-methyl-2 ''-butenyl)chalcone, 1] is a prenylated chalcone that has broad biological activity, in particular against neuroblastomas, the most common cancer in infancy. It is currently commercially available at a cost of $190/mg by extraction from Psoralea corylifolia and a number of other African and Asian plants. Several synthetic routes have been explored, and the most efficient procedure involves the palladium-catalysed Stille coupling of 3-iodo-2,4-bis(methoxymethoxy)acetophenone (25) with prenyltributyltin, Claisen-Schmidt condensation with 4-(methoxymethoxy)benzaldehyde to form the triply MOM-protected prenylchalcone 27 and finally deprotection with 2 M HCl in methanol to form isobavachalcone in an overall yield of 15% over five steps. The X-ray crystal structures of 2,4-dihydroxy-3-iodoacetophenone (21) and of several prenylated chalcones are reported, including the elucidation of their hydrogen-bonding networks in the solid state. The synthetic route has been extended to include organometallic derivatives in which the 4-(methoxymethoxy) benzaldehyde used in the Claisen-Schmidt condensation has been replaced by formylferrocene, formylruthenocene or (eta(5)-formylcyclopentadienyl)(eta(4)-tetraphenylcyclobutadiene)cobalt to form the corresponding analogues of isobavachalcone containing organometallic sandwich moieties.

Summary: Organometallic quinone methides (OM-QMs) exhibit unique chemical and biological properties when incorporated into specific substrates like ferrocifens, leading to disruption of cancer cell metabolism and cytotoxicity. Among these, ferrocifens show the highest toxicity within the iron, ruthenium, osmium triad, and the complexes can be utilized as imaging probes.

Dynamics of a Molecular Rotor Exhibiting Local Directional Rotational Preference within Each Enantiomer

Summary: Directional internal rotation in molecular systems is typically controlled by chirality, and a designed molecular rotor with ferrocene and triptycene linked by a methylene bridge provides two rotational degrees of freedom. Experimental observations and computational data suggest strong coupling between the two rotation modes, with the spatial orientation of the bistable ferrocene moiety influencing its rotation barrier. These findings demonstrate the potential for locally preferred thermal directional intramolecular rotation in dynamically interconverting enantiomers.

Diversity-oriented synthesis and bioactivity evaluation of N-substituted ferrocifen compounds as novel antiproliferative agents against TNBC cancer cells

Summary: Ferrociphenols, compounds characterized by a biologically active redox motif, have anticancer properties. This study synthesized a variety of ferrociphenols with nitrogen-containing substituents and investigated their structure-activity relationships. Oxidative metabolism experiments and ROS-related bioactivity measurements were conducted to explore the cytotoxicity of imidoferrociphenols. Additionally, dimerization phenomenon observed in the crystal structure of a ferrociphenol may affect its antitumor activity.

Intercluster exchanges leading to hydride-centered bimetallic clusters: a multi-NMR, X-ray crystallographic, and DFT study

Summary: Encouraged by successful syntheses of alloy nanoclusters through intercluster reactions, a series of bimetallic hydride clusters were prepared and characterized using ESI-MS, multi-NMR, and X-ray diffraction. Metal exchanges in solution drove a dynamic chemical equilibrium, as indicated by P-31 NMR spectroscopy and 2D exchange spectroscopy. DFT calculations confirmed the preferential metal positions within the clusters, as observed in crystal structures.
