Influence of alcohol on the release of tramadol from 24-h controlled-release formulations during in vitro dissolution experiments

Recent warnings by regulatory bodies and a product recall by the FDA have generated much interest in the area of dose dumping from controlled-release opioid analgesic formulations when coingested with alcohol. It was the aim of this study to address this issue and in doing so, gain understanding on how alcohol-induced effects may be avoided. In this study, tramadol release from Ultram (R) ER tablets and T-long (R) capsules was significantly increased in the presence of ethanol. Conversely, a decrease in the rate of tramadol release was seen from Tridural (TM) extended-release tablets in the presence of alcohol.

Increasing Cellular Uptake and Permeation of Curcumin Using a Novel Polymer-Surfactant Formulation

Summary: This study investigated the potential of novel polymer-surfactant-based formulations to improve the solubility and permeability of curcumin. Engineering artesunate-loaded micelles using spray drying for pulmonary drug delivery

Summary: Novel dry powder formulations of D-α-tocopheryl polyethylene glycol 1000 succinate (TPGS) micelles containing the anti-cancer drug artesunate (ART) were developed for inhalation therapy in lung cancer treatment. The study successfully prepared TPGS micelles encapsulating ART using thin-film hydration method and then transformed them into dry powder formulations through spray-drying with lactose and leucine as carrier molecules. Characterization of the dry powder formulation showed a unimodal particle size distribution with a mean particle size of 3.27 μm and a fine particle fraction of 31%. The artesunate-loaded micelle dry powder formulation exhibited higher cytotoxicity against A549 lung cancer cells compared to docetaxel and doxorubicin. This study demonstrates the potential of spray-dried micelles containing artesunate as a dry powder formulation for inhalation therapy in lung cancer.
