The value of combined use of survivin mRNA and matrix metalloproteinase 2 and 9 for bladder cancer detection in voided urine

Objective: In a trial to improve the diagnostic efficacy of conventional urine cytology we determine survivin RNA and matrix metalloproteinase 2 and 9 in urine of bladder cancer cases. Method: Voided urine specimens were collected from patients with histologically confirmed bladder urothelial carcinoma (Group 1; n = 46), urological patients without urothelial carcinoma (Group 2; n = 20), and healthy volunteers (Group 3; n = 20). Urine cytology, survivin RNA was estimated by qualitative nested RT-PCR and MMP-2, MMP-9 activity were detected by gelatin zymography. The expression of survivin RNA and matrix metalloproteinase 2 and 9 in bladder cancer was compared with benign and normal cases. Results: Positivity rates of survivin RNA and MMPs zymography were significantly different among the 3 groups. Urine survivin detection by qualitative nested RT-PCR showed 76.1% sensitivity and 95% specificity. The overall sensitivity, specificity of urinary MMP zymography was 67.3%, 90% respectively. The combined use of urine cytology with urine survivin or MMPs zymography increased sensitivity of urine cytology from 50% to 84.7%. The highest sensitivity (95.6%) was obtained on combining the three markers. Conclusion: Survivin RNA and MMPS zymography can be considered as promising noninvasive markers for bladder cancer early detection. Combined use of the three markers improved the sensitivity for detecting bladder cancer.

This provides valuable insights for potential diagnostic and management strategies for NAFLD.

Summary: This study analyzed the impact and distribution of blood groups in different ethnicities and the susceptibility to COVID-19 infection in Makkah, Saudi Arabia. The results showed that blood types A and O were associated with higher infection rates, while blood type AB was associated with lower infection rates. Blood types A and O play a critical role in COVID-19 prognosis and vaccination priority.

Body mass index is associated with Helicobacter pylori infection and increased oxidative DNA damage in an obese population

Summary: The objective of this study was to determine the prevalence of Helicobacter pylori infection in a Saudi Arabian population and investigate its association with body mass index (BMI) and serum 8-hydroxy deoxyguanine (8-OHdG) levels. The results showed that obese individuals had a higher prevalence of H. pylori infection, and the levels of 8-OHdG were positively correlated with BMI in H. pylori-positive patients.

Modulation of hepatic stellate cells by Mutaflor® probiotic in non-alcoholic fatty liver disease management

Summary: This study constructed a genetic-epigenetic network linked to hepatic stellate cell (HSC) signaling and evaluated the effects of Mutaflor (R) on this network. The results showed that Mutaflor (R) administration minimized the alterations caused by NAFLD/NASH and reduced HSC activation and profibrogenic protein expression. This effect may be achieved through upregulation of RPARP AS-1 LncRNA and downregulation of miR-650, FOXA2 mRNA, and TEAD2 mRNA expression.

Evaluation of the role of kefir in management of non-alcoholic steatohepatitis rat model via modulation of NASH linked mRNA-miRNA panel

Summary: Non-alcoholic steatohepatitis (NASH), the aggressive variant of non-alcoholic fatty liver disease, can be regulated through the Hippo pathway. Probiotics, such as kefir milk, have been shown to effectively manage NASH. In this study, the efficacy of kefir milk in NASH management was investigated through the modulation of hepatic mRNA-miRNA panel linked to NAFLD/NASH Hippo signaling and gut microbita regulated genes. The results showed that kefir administration prevented steatosis and inflammatory component of NASH, improved liver function and lipid panel, and down-regulated the expression of miR 6807-5p while up-regulating the expression of SOX 11 and AMOTL2, resulting in the reduction of inflammatory and fibrotic markers. Overall, kefir suppressed NASH progression and improved fibrosis and hepatic inflammation.

Investigation of Hyperlipidemia Associated with Increased Levels of Oxidized Low-Density Lipoproteins and 8-Hydroxy-2 acute accent -Deoxyguanosine

Summary: The aim of this study was to evaluate the association between lipid status and the levels of oxidized low-density lipoproteins and 8-hydroxy-2'-deoxyguanosine. The results showed a positive correlation between oxidized low-density lipoproteins and 8-hydroxy-2'-deoxyguanosine levels with hypercholesterolemia, hypertriglyceridemia, and high levels of low-density lipoproteins. This suggests that disturbance in lipid profile may result in increased levels of oxidized low-density lipoproteins and oxidative stress.
