Aim: The main goal of the present study was the evaluation of ischemia-modified albumin (IMA) in patients with type 2 diabetes mellitus and estimation of its connection with vascular complications, glycemic control, hypertension, dyslipidemia and obesity. Methods: In 76 diabetic patients and 25 control subjects, a plasma level of IMA by manually performed, spectrophotometric Co(II)-albumin binding assay was determined. Other parameters such as glucose, fructosamine, HbA(1c), total cholesterol and its fractions (HDL, LDL), triglicerydes were estimated by routine methods. Results: Diabetic patients had significantly higher level of IMA in comparison with control subjects. There were not significant differences between groups with various states of vascular complications although the lowest concentration of IMA was observed in patients with microangiopathy. Patients with poor glycemic control had higher IMA level in comparison with these with good glycemic control. Significant correlation was observed between IMA and HbA1c. Among the risk factors, only blood pressure and LDL showed a weak relationship with IMA level. Conclusions: Our results revealed, for the first time, higher level of IMA in diabetic patients which confirms that it may be of non-cardiac origin. We can suggest that the albumin molecule in plasma of diabetic patients is modified in the chronic hypoxia conditions provoked mainly by hyperglycemia and oxidative stress in diabetes.

Summary: The role of medical personnel in promoting vaccination and pro-health attitudes is crucial for protection against COVID-19. A study assessed the attitudes of healthcare workers and medical students towards preventive vaccinations against COVID-19 and found that young medical students had a more positive attitude, while students expressed higher concerns about the vaccine's side effects and contracting the virus.

Summary: The aim of this study was to evaluate and compare the effectiveness of different vaccines in Poland's mass vaccination program and investigate the most common side effects. The study found that 100% of patients responded to the vaccines, with convalescents having higher antibody levels after the first dose but a decrease after the second dose. Moderna vaccine generated the highest antibody titers after both doses, and fever was identified as a significant factor in higher antibody levels.

Evaluation of Pro/Antioxidant Imbalance in Blood of Women with Polycystic Ovary Syndrome Based on Determination of Oxidized Low-Density Lipoproteins and Ferric Reducing Ability of Plasma Values

Justyna Niepsuj, Grzegorz Franik, Pawel Madej, Agnieszka Piwowar, Anna Bizon

Summary: In women with polycystic ovary syndrome (PCOS), factors such as body mass index, waist-to-hip ratio, age, and glucose concentration significantly affect oxLDL-C and FRAP levels. Furthermore, there is a correlation between FRAP and oxLDL-C levels in the whole group and in insulin-sensitive women. Additionally, the concentration of Anti-Mullerian hormone is negatively related to oxLDL-C and FRAP levels.

Summary: This study reviews the role of SIRTs in the physiology and pathology of the central nervous system (CNS) and discusses their crucial involvement in neuroinflammation, neurodegeneration, and metabolic alterations. The understanding of SIRTs' role in various CNS diseases provides an opportunity to identify relevant markers of disease progression and potential therapeutic targets.

The Effect of Metalloestrogens on the Effectiveness of Aromatase Inhibitors in a Hormone-Dependent Breast Cancer Cell Model

Kamila Boszkiewicz, Helena Moreira, Ewa Sawicka, Anna Szyjka, Agnieszka Piwowar

Summary: Progressive industrialization, urbanization, and consumerism have led to increased contamination of the environment with endocrine-disrupting compounds (EDCs), which play a crucial role in the rise of hormone-dependent cancers, including breast cancer. This study aimed to investigate the impact of exposure to specific xenoestrogens, commonly found in daily life (aluminum in antiperspirants and deodorants; chromium (III) in dietary supplements and drugs), on the effectiveness of hormone therapy in breast cancer treatment. In vitro tests were conducted on breast cancer cell lines exposed to xenoestrogens, drugs, and their combinations, and the results showed that such exposure can impair the efficacy of hormone therapy.

Evaluation of Selected Oxidant/Antioxidant Parameters in Patients with Relapsing-Remitting Multiple Sclerosis Undergoing Disease-Modifying Therapies

Anna Bizon, Justyna Chojdak-Lukasiewicz, Aleksandra Koltuniuk, Slawomir Budrewicz, Anna Pokryszko-Dragan, Agnieszka Piwowar

Summary: This study evaluated oxidative stress parameters in the serum of patients with relapsing-remitting multiple sclerosis (RRMS), and found associations between these parameters and clinical/laboratory markers. AOPP and FRAP concentrations were higher in male patients. There were correlations between AOPP and inflammation markers, and differences in AOPP among subgroups receiving different treatments.

Monika Stepien, Natalia Swiatoniowska-Lonc, Brygida Knysz, Beata Jankowska-Polanska, Amadeusz Kuzniarski, Agnieszka Piwowar, Malgorzata Zalewska

Summary: This study aimed to evaluate the levels of anti-SARS-CoV-2 immunoglobulin G (IgG) antibodies in convalescents and individuals without a past infection, and to assess socio-clinical factors associated with participants' humoral responses. The results showed the presence of anti-SARS-CoV-2 IgG antibodies in convalescents and in some individuals without a past infection, and the antibody levels were related to the severity of the symptoms.

HIGH GLUCOSE REDUCES THE ANTI-TUMOR ACTIVITY OF AROMATASE INHIBITORS IN A HORMONE-DEPENDENT BREAST CANCER CELL MODEL

Kamila Boszkiewicz, Agnieszka Piwowar

Summary: One-third of breast cancer patients with diabetes have a 40% higher risk of mortality. Metalloestrogens may reduce the effectiveness of aromatase inhibitors (AIs), but high glucose conditions have a greater impact on their activity. Therefore, controlling hyperglycemia and individualized treatment regimens may have important therapeutic implications for cancer patients with diabetes.

Exploring the Relationship between Antioxidant Enzymes, Oxidative Stress Markers, and Clinical Profile in Relapsing-Remitting Multiple Sclerosis

Anna Bizon, Justyna Chojdak-Lukasiewicz, Slawomir Budrewicz, Anna Pokryszko-Dragan, Agnieszka Piwowar

Summary: This study aimed to investigate the alterations in the antioxidant balance in patients with RRMS and the factors influencing it. The study included 161 patients and 29 healthy individuals, and measured the activity of SOD, GPx, and CAT, as well as the concentration of IL-6, LPO, TOS, and TAS. The results showed decreased GPx activity and increased CAT activity in patients with RRMS compared to the control group, and these activities were influenced by gender and disease-modifying therapies. The findings suggest that enhancing GPx activity may be a potential therapeutic strategy for mitigating oxidative stress in RRMS.

EXPRESSION OF PARAOXONASE 1 IN HEK293 CELLS AND ITS EFFECT ON LACTONASE AND ARYLESTERASE ACTIVITY IN SAMPLES CONTAINING HUMAN SERUM AND/OR PROOXIDANT OR ANTIOXIDANT AGENTS- A PRELIMINARY STUDY

Anna Bizon, Agnieszka Piwowar

Summary: The expression of recombinant paraoxonase 1 (PON1) in the human embryonic kidney (HEK293F) cell line was investigated. It was demonstrated that the HEK293F platform is useful for obtaining recombinant human PON1 protein. The addition of high-density lipoprotein increased serum arylesterase and lactonase activity of PON1, while low-density lipoprotein, copper ions, and iron ions decreased PON1 activity.
