Summary: Mesenchymal stromal cells (MSCs) have been shown to play a supportive role in the expansion and engraftment of hematopoietic stem and progenitor cells (HSPCs). In this study, we developed a co-culture system using bone marrow (BM)-MSCs to improve the transplantation outcome of CRISPR-Cas9 gene-edited (GE) human HSPCs. We found that BM-MSCs can produce hematopoietic supportive and anti-inflammatory factors that enhance the expansion and clonogenic potential of GE-HSPCs in vitro, leading to improved engraftment and early phase hematological reconstitution in transplanted mice. This study suggests a potential clinical application of BM-MSCs in promoting successful transplantation of GE-HSPCs.

Summary: In a rat calvarial defect model, adding endothelial cells (ECs) to a culture of bone marrow stromal cells (BMSCs) has been reported to enhance bone formation significantly. This study aimed to further investigate the ossification process of newly formed osteoid and host response to poly(L-lactide-co-1,5-dioxepan-2-one) [poly(LLA-co-DXO)] scaffolds. The results showed increased expression of endochondral ossification-related genes in the CO group, along with the formation of cartilage-like tissue in the scaffold pores and the observation of multinucleated giant cells (MNGCs) surrounding the scaffold fragments.
