Purpose of review Increasing use of drugs among elderly people has raised concerns about possible negative health outcomes, including malnutrition, associated with polypharmacy. Evidence about the association of polypharmacy with nutritional status is scarce. This review summarizes the relevant evidence regarding polypharmacy and nutritional status in elderly people. Recent findings The probability of nutritional problems as a consequence of drugs is highest in elderly people suffering from several diseases. Drug treatment may contribute to poor nutritional status by causing loss of appetite, gastrointestinal problems, and other alterations in body function. Some recently published studies add evidence on possible association between increasing number of drugs and malnutrition. Studies indicate also an association between polypharmacy and weight changes. In addition, there are available studies that have shown deficits in the intake of specific macronutrients and micronutrients (e.g. fiber, glucose, and specific vitamins) for those with a high number of drugs in use. Summary On the basis of available evidence, the role of polypharmacy on nutritional status among elderly people is unclear. Some diseases promote malnutrition; thus, the independent role of drugs for nutritional status is challenging to determine. Longitudinal studies with careful adjustment for underlying diseases are needed to explore association between polypharmacy and malnutrition. Nutritional evaluation should be a routine part of comprehensive geriatric assessment that is conducted ideally in multiprofessional teams, including physician, pharmacist, and dietitian.

P2Y2 receptor activation promotes esophageal cancer cells proliferation via ERK1/2 pathway

Aline Zaparte, Angelica R. Cappellari, Caroline A. Brandao, Julia B. de Souza, Thiago J. Borges, Luiza W. Kist, Mauricio R. Bogo, Luiz F. Zerbini, Luis Felipe Ribeiro Pinto, Talita Glaser, Maria Carolina B. Goncalves, Yahaira Naaldijk, Henning Ulrich, Fernanda B. Morrone

Summary: Esophageal cancer is a global disease with two main subtypes, esophageal squamous cell carcinoma and esophageal adenocarcinoma. Research shows that P2Y(2) receptor activation in esophageal cancer cells promotes cell proliferation and colony formation, while its blockade reduces these effects. Targeting P2Y(2) receptors may have therapeutic potential for esophageal cancer treatment.

Immunosuppression in Gliomas via PD-1/PD-L1 Axis and Adenosine Pathway

Thamiris Becker Scheffel, Nathalia Grave, Pedro Vargas, Fernando Mendonca Diz, Liliana Rockenbach, Fernanda Bueno Morrone

Summary: Glioblastoma is a highly malignant subtype of glioma, and the immune landscape within the tumor microenvironment plays a crucial role in influencing therapy outcomes. The presence of multiple immunosuppression pathways, particularly the adenosine pathway, poses significant obstacles to immune-based cancer therapies and can contribute to treatment resistance. Research focusing on immune checkpoints and the adenosine pathway may provide insights into potential targets for improving tumor treatment strategies.

Morphological and mechanical changes induced by quercetin in human T24 bladder cancer cells

Bruno Silveira Adami, Fernando Mendonca Diz, Gustavo Petry Oliveira Goncalves, Camille Kirinus Reghelin, Matheus Scherer, Artur Pereira Dutra, Ricardo Meurer Papaleo, Jarbas Rodrigues de Oliveira, Fernanda Bueno Morrone, Andrea Wieck, Leder Leal Xavier

Summary: The study demonstrates that quercetin inhibits proliferation and induces apoptosis and senescence in bladder cancer cells. It also causes morphological and mechanical alterations in the cells, including decreased cell viability, cytoplasmic retraction, and membrane condensation. The findings suggest a potential anticancer effect of quercetin through its impact on cellular morphology and mechanics.

Pannexin channel 1, P2x7 receptors, and Dimethyl Sulfoxide mediate pain responses in zebrafish

Darlan Gusso, Fernanda Fernandes Cruz, Pamella Moreira Fritsch, Marilia Oberto Gobbo, Fernanda Bueno Morrone, Carla Denise Bonan

Summary: This study evaluated the perceived pain in zebrafish larvae by observing behavioral changes. The results showed that acetic acid induced behavioral changes in zebrafish larvae, and compounds such as ethanol, ibuprofen, and paracetamol could prevent these changes. In addition, antagonists and inhibitors of specific channels also prevented the behavioral changes induced by acetic acid. These findings suggest that zebrafish can serve as a translational model for studying pain, as their early developmental stages exhibit behavioral responses similar to mammals.

High P2X6 receptor expression in human bladder cancer predicts good survival prognosis

Fabricia Dietrich, Angelica Regina Cappellari, Eduardo Cremonese Filippi-Chiela, Patricia Boni de Paula, Julia Brandt de Souza, Stefano Walter Agatti, Roberta Andrejew, Rafael Roesler, Fernanda Bueno Morrone, Ana Maria Oliveira Battastini

Summary: Through clinical data and in vitro experiments, we found that the P2X6 receptor plays a prognostic role in bladder cancer. High expression of P2X6 receptor is associated with increased survival rate in patients. After radiotherapy, P2X6 receptor expression is upregulated in T24 cells but not in RT4 cells.

Summary: This study investigated the role of the P2Y(12) receptor in the proliferation and migration of GBM cells and evaluated its expression in patients' data. The results showed that the P2Y(12) antagonist, ticagrelor, decreased the viability and proliferation of GBM cells and reduced colony formation and migration potentials. The expression of P2Y(12)R was found to be higher in glioma samples, suggesting its importance in glioma development.

The functional role of p38 MAPK pathway in malignant brain tumors

Nathalia Grave, Thamiris Becker Scheffel, Fernanda Fernandes Cruz, Liliana Rockenbach, Marcia Ines Goettert, Stefan Laufer, Fernanda Bueno Morrone

Summary: Gliomas are debilitating malignant brain tumors with limited response to therapies, driven by molecular abnormalities like mutations in regulatory networks. The MAPK pathway, particularly p38 MAPK activation, plays a crucial role in glioma cell invasion and metastasis, and is associated with tumor grade and chemotherapy resistance, underlining its potential as a therapeutic target.
