Polarity within lymphocytes has been recognized to regulate a variety of processes, including migration, signaling, and the execution of effector function. It has been recently proposed, however, that this polarized behavior may also serve a different purpose in lymphocytes that have not yet encountered their foreign antigen - to coordinate asymmetric cell division. Asymmetric division is an evolutionarily conserved mechanism allowing a single cell to give rise to two distinct daughter cells from inception. In this review, recent findings in polarity and asymmetric division in lymphocytes are discussed.

Summary: This study assessed the differences in SARS-CoV-2 vaccine immunogenicity between patients with IBD treated with different immune-modulating medications and normal healthy donors. The results showed reduced humoral immunity in patients with IBD on infliximab, but no difference in humoral and cell-mediated immunity in patients with IBD on infliximab with a thiopurine or vedolizumab compared with normal healthy donors.

Summary: This study investigates the diversity of gene expression and genome accessibility in mouse CD8(+) T-RM cells in different tissues, and identifies molecules critical for the generation of CD8(+) T-RM cells in response to acute viral infection. The research reveals both common and tissue-specific transcriptional and epigenetic signatures in T-RM cells from different organs. T-RM cells in the intestine and salivary glands express TGF-beta-induced genes and are maintained by ongoing TGF-beta signaling, while those in the fat, kidney, and liver do not. Furthermore, the study identifies the transcriptional repressor Hic1 as a critical regulator of T-RM cell differentiation in the small intestine, and shows that Hic1 overexpression enhances T-RM cell differentiation and protection from infection.

Summary: Tissue-resident memory CD8+ T (TRM) cells are a subset of memory T cells that are important for controlling infection. TRM cells in the small intestine and colon exhibit heterogeneity in terms of cytokine and granzyme expression, transcriptional and epigenetic regulation, and functional characteristics. The transcription factor Eomes plays a context-specific role in supporting the maintenance of established TRM cells in the small intestine but not in the colon.

Summary: This study investigated the molecular mechanisms of hyperbaric oxygen therapy (HBOT) for moderate-to-severe ulcerative colitis. The findings indicate that HBOT reduces neutrophil STAT3 and azurophilic granule activity in UC patients, and improves colitis activity by altering microbial composition and metabolism. The strain-level variations in A muciniphila in the intestinal microbiota may contribute to the nonresponse to HBOT.

Skin inflammation activates intestinal stromal fibroblasts and promotes colitis

Summary: This study revealed that intestinal inflammation can promote the differentiation of intestinal stromal fibroblasts into preadipocytes with innate antimicrobial activity, and this process may be exacerbated by skin inflammation in mice. The activation of hyaluronan catabolism during skin injury was found to alter colon fibroblast function and promote excessive reactive adipogenesis, linking skin and colon inflammation through the release of hyaluronan.

Distinct integrin activation pathways for effector and regulatory T cell trafficking and function

Summary: Integrin activation is crucial for lymphocyte trafficking and immune function. RIAM is necessary for conventional T cell integrin activation, but dispensable for regulatory T cell integrin activation and suppressive function. Inhibition of RIAM can prevent spontaneous Tconv cell-mediated autoimmune colitis while maintaining T reg cell trafficking and function.
