Targeting the Akt Kinase to Modulate Survival, Invasiveness and Drug Resistance of Cancer Cells

The deregulation of oncogenic signaling pathways which provide survival advantages to tumor cells is mediated by multiple cellular networks. Among them, the PI3K-Akt-mTOR axis, in particular the serine/threonine kinase Akt, is recognized as a key player. The kinase is hyperactivated due to a variety of mechanisms including loss of PTEN, mutations in the PI3K catalytic subunit, receptor tyrosine kinase and Ras activation. Indeed, inappropriate activation of the Akt kinase is a common event in human tumors and Akt appears to be a critical player in cell survival that may also account for the therapeutic resistance and the invasive phenotype of tumors. Inhibition of Akt signalling results in apoptosis and growth inhibition of tumour cells with elevated Akt activity. A functional role in drug resistance is supported by evidence that tumor cells with acquired resistance to antitumor agents may display increased Akt activation and that treatment with molecularly targeted agents can activate feed-back loops involving Akt. This serine/threonine kinase may therefore represent an amenable target for modulation of sensitivity to compounds with different molecular features due to its pleiotropic role in cell survival. Different types of Akt inhibitors [i.e., ATP mimetics and pleckstrin-homology (PH) domain binders] have been generated and some of them have reached the clinical setting. The present review focuses on the i) mechanisms implicating Akt in increased survival and invasive potential of tumor cells of different tumor types and ii) on the development of Akt inhibitors as modulators of drug resistance.

Increase of Circulating Endothelial Progenitor Cells and Released Angiogenic Factors in Children with Moyamoya Arteriopathy

Summary: This study aimed to assess the levels of circulating endothelial progenitor cells (cEPC) and the release of selected proteins in the plasma and CSF of pediatric MMA patients to clarify the pathogenesis and progression of the disease. The study found increased levels of cEPC in peripheral blood and an upregulation of angiogenic markers in CSF. These findings provide valuable insights into the disease's mechanism and progression.

The Therapeutic Potential of Pyroptosis in Melanoma

Summary: Pyroptosis is a programmed cell death characterized by the rupture of the plasma membranes and release of cellular content leading to inflammatory reaction. Four cellular mechanisms inducing pyroptosis have been reported thus far. Pyroptosis plays roles in tumor initiation, progression, metastasis, treatment response, and patient outcome. Induction of pyroptosis has been reported in several tumor types following antitumor therapies, including melanoma.

Summary: Peripheral nerve sheath tumors (PNST) mainly include schwannomas and neurofibromas. Surgical resection is the primary treatment, but distinguishing between intact functional nerve and the fibers from which the PNST arose can be challenging. Intraoperative tools to visualize these tumors could improve the success rate of gross-total resection.

Effectiveness of irinotecan plus trabectedin on a desmoplastic small round cell tumor patient-derived xenograft

Summary: This study used a patient-derived xenograft (PDX) model to evaluate the effectiveness of different drugs against desmoplastic small round cell tumor (DSRCT). It was found that doxorubicin, pazopanib, and larotrectenib had moderate antitumor effects, while trabectedin had higher activity. Vinorelbine, irinotecan, and eribulin achieved nearly complete tumor growth inhibition. The combination of irinotecan with either eribulin or trabectedin resulted in complete responses. This study highlights the importance of patient-derived preclinical models in exploring new treatments for DSRCT.

Elagolix Sodium Salt and Its Synthetic Intermediates: A Spectroscopic, Crystallographic, and Conformational Study

Summary: Elagolix sodium salt is the first orally active non-peptide gonadotropin-releasing hormone receptor antagonist for the treatment of hormone dependent diseases. A detailed NMR investigation was conducted to analyze the drug and its synthetic intermediates, allowing for the complete assignment of the NMR signals and determination of the stereochemical profile of the atropisomers. These results contribute to the understanding of atropisomerism in drug discovery and can be applied in the design of safer and more effective analogs.

Summary: Moyamoya angiopathy (MMA) is a rare cerebrovascular disease characterized by progressive steno-occlusive lesions and collateral vessel development. Surgical treatment, particularly indirect revascularization, is preferred. By performing proteomic characterization, a potential gene, Filamin A, was identified, suggesting a role in MMA pathogenesis. This study provides insights into the molecular understanding of this disabling intracranial vasculopathy.

Synthesis of N-oxyamide analogues of protein kinase B (Akt) targeting anionic glycoglycerolipids and their antiproliferative activity on human ovarian carcinoma cells

Summary: Bioactive anionic glycoglycerolipids based on 2-O-& beta;-d-glucosylglycerol were efficiently synthesized using N-oxyamides. The oxidation of the glucose moiety's primary hydroxyl group in the presence of N-oxyamide was achieved for the first time using a chemoenzymatic laccase/TEMPO procedure. The N-oxyamides showed higher inhibition of ovarian carcinoma IGROV-1 cell proliferation in serum-free medium, indicating increased antiproliferative activity compared to the corresponding esters.
