Potential Role of the Mitochondria for the Dermatological Treatment of Papillon-Lefevre

Summary: Papillon-Lefevre syndrome is a rare genetic disorder characterized by periodontitis and palmoplantar hyperkeratosis, with main inflammatory deficiencies in oxidative stress and autophagic dysfunction. Mitochondrial dysfunction has been found to be associated with the syndrome, which may also lead to a decrease in oxygen consumption rate. CoQ10 supplementation could improve mitochondrial function and potentially serve as a local treatment option for patients with this syndrome.

Summary: This study showed increased expression of NLRP3 inflammasome components in HGPS skin fibroblasts and lymphoblasts, as well as in hearts and livers of Zmpste24(-/-) mice. Inhibiting the NLRP3 inflammasome with MCC950 improved cellular phenotype, extended lifespan of progeroid animals, and reduced inflammation, suggesting a potential therapeutic approach for HGPS.

Summary: The study revealed impaired autophagy and increased lysosomal permeability in skin fibroblasts obtained from patients with Tay-Sachs and Sandhoff diseases. Additionally, it was found that the mTOR signaling pathway was compromised, but supplementation of L-arginine partially restored mTOR activity and improved cellular abnormalities.

Summary: In the past, mtROS were seen as byproducts of cellular metabolism and drivers of ageing and age-related diseases. Now, we know that mtROS serve as important cellular messengers, playing a role in maintaining cellular homeostasis and influencing cellular differentiation, proliferation, and survival. However, dysregulated mtROS signaling can contribute to degenerative diseases. This review focuses on the signaling pathways involving mtROS and their role in ageing.
