Effect of dark chocolate on renal tissue oxygenation as measured by BOLD-MRI in healthy volunteers

Background: Cocoa is rich in flavonoids, has anti-oxidative properties and increases the bioavailability of nitric oxide (NO). Adequate renal tissue oxygenation is crucial for the maintenance of renal function. The goal of this study was to investigate the effect of cocoa-rich dark chocolate (DC) on renal tissue oxygenation in humans, as compared to flavonoid-poor white chocolate (WC). Methods: Ten healthy volunteers with preserved kidney function (mean age +/- SD 35 +/- 12 years, 70% women, BMI 21 +/- 3 kg/m(2)) underwent blood oxygenation level-dependent magnetic resonance imaging (BOLD-MRI) before and 2 hours after the ingestion of 1 g/kg of DC (70% cocoa). Renal tissue oxygenation was determined by the measurement of R2* maps on 4 coronal slices covering both kidneys. The mean R2* (= 1/T2*) values in the medulla and cortex were calculated, a low R2* indicating high tissue oxygenation. Eight participants also underwent BOLD-MRI at least 1 week later, before and 2 hours after the intake of 1 g/kg WC. Results: The mean medullary R2* was lower after DC intake compared to baseline (28.2 +/- 1.3 s(-1) vs. 29.6 +/- 1.3 s(-1), p = 0.04), whereas cortical and medullary R2* values did not change after WC intake. The change in medullary R2* correlated with the level of circulating (epi)catechines, metabolites of flavonoids (r = 0.74, p = 0.037), and was independent of plasma renin activity. Conclusion: This study suggests for the first time an increase of renal medullary oxygenation after intake of dark chocolate. Whether this is linked to flavonoid-induced changes in renal perfusion or oxygen consumption, and whether cocoa has potentially renoprotective properties, merits further study.

Focused navigation for respiratory-motion-corrected free-running radial 4D flow MRI

Summary: The study validated a respiratory motion correction method called focused navigation (fNAV) for free-running radial whole-heart 4D flow MRI. Using fNAV, respiratory signals were converted into three orthogonal displacements to correct respiratory motion. The difference between generated and fNAV displacement coefficients was calculated and compared to the motion-free ground-truth. Results showed that fNAV corrected respiratory motion and improved 4D flow measurements compared to uncorrected datasets.

Summary: This study aimed to investigate the dose-response effect of hydrochlorothiazide agents for preventing the recurrence of kidney stones. The results showed that there was no significant difference in the incidence of recurrent stones among patients receiving hydrochlorothiazide at doses of 12.5 mg, 25 mg, or 50 mg compared to placebo. However, the use of hydrochlorothiazide was associated with a higher risk of adverse reactions.

Determinants of Renal Micro-Perfusion as Assessed with Contrast-Enhanced Ultrasound in Healthy Males and Females

Summary: In this study, contrast-enhanced ultrasound was used to quantitatively evaluate the cortical micro-perfusion, and it was found that females had higher cortical micro-perfusion index than males, but this difference was no longer significant after adjusting for covariates. Applications of the Microscale Thermophoresis Binding Assay in COVID-19 Research

Summary: As new variants of SARS-CoV-2 continue to emerge during the COVID-19 pandemic, it is crucial to develop optimized tools for studying these variants and potential future coronaviruses. This report introduces microscale thermophoresis (MST) as a reliable and versatile tool for coronavirus research. The study demonstrates three applications of MST, including the binding of SARS-CoV-2 spike receptor binding domain (RBD) to peptides, the binding of RBD to the viral receptor ACE2, and the binding of RBD to ACE2 in complex with the amino acid transporter SLC6A20/SIT1 or its allelic variant rs61731475 (p.Ile529Val). The results highlight MST as a highly precise approach for studying protein interactions and developing antiviral agents in coronavirus research, particularly its advantage in measuring interactions with membrane proteins in their near-native plasma membrane environment.

Efficacy and safety of avacopan in patients with ANCA-associated vasculitis receiving rituximab in a randomised trial

Summary: This study evaluated the efficacy and safety of avacopan in a subgroup of patients with antineutrophil cytoplasmic autoantibody (ANCA)-associated vasculitis receiving rituximab as background induction therapy. The results showed that avacopan was comparable to prednisone taper in achieving remission at week 26, but had greater efficacy at week 52. Additionally, avacopan was associated with improved renal outcomes and lower glucocorticoid toxicity.
