Summary: A fast, simple, and reliable LC-MS/MS method has been developed and validated for the determination of 24-hour urinary 5-HIAA, and the quality assurance for post-implementation monitoring has been explored. The reference interval of 24-hour urinary 5HIAA in Chinese adults has been established, ensuring reliable and comparable results in routine clinical testing. This LC-MS/MS method can be proposed as a clinical testing tool for urinary 5-HIAA in quality-controlled environments.

Summary: Researchers have successfully synthesized a double-mechanism-driven fluorescent sensor based on a Tb-based metal-organic framework (Tb-DBA), which can quantitatively recognize urinary vanillylmandelic acid (VMA) as an early pathological signature of neuroblastoma (NEURO) using two completely different luminescence phenomena in a smaller concentration window by switching the excitation wavelength. This work represents the first example of designing a luminescent sensor coupled with multiple mechanisms in a single sensing platform for the diagnosis of NEURO, providing a new avenue for the clinical application of two-dimensional detection biomarkers for accurate diagnosis in serious diseases.

Summary: In this report, the first voltammetric sensor for simultaneous determination of neuroendocrine tumour biomarkers 5-Hydroxy indole acetic acid (HIAA) and Vannillyl mandelic acid (VMA) is presented. A glassy carbon electrode modified with a copolymer of taurine and glutamic acid was used to detect the analytes. The combined effect of the two conducting polymers showed significant voltammetric response for the biomarkers. Calibration measurements were conducted using a fast and feasible square wave voltammetric technique. The voltammetric sensor achieved detection limits as low as 4.19 x 10(-8) M (HIAA) and 6.16 x 10(-8) M (VMA) with tolerable interference from coexisting species. Additionally, spike recovery analysis of the analytes in artificial urine samples showed adequate recovery values, demonstrating the reliability of the sensor.

Calibration graphs were linear for HVA within a certain concentration range, and simultaneous assay of both compounds showed acceptable average recovery. This study lays the foundation for simultaneous quantification of these compounds in urine samples for Neuroblastoma diagnosis.

Summary: In this research, a single-component multi-colorimetric sensor array was developed to simultaneously detect homovanillic acid (HVA) and vanillylmandelic acid (VMA) tumor markers. Gold nanorods (AuNRs) were used as optical signal transducers, and the aspect ratio alterations of AuNRs were achieved by silver shell metallization. The sensor array showed the potential to diagnose childhood metabolic errors with 100% sensitivity and specificity in detecting HVA, VMA, and HVA:VMA in human urine samples.

However, there are challenges in using these tests in children, and the true prevalence of childhood PPGL may be underestimated. Early identification and accurate diagnosis are important for improving therapeutic outcomes.

Summary: This observational study aimed to develop a new LC-MS/MS method for TDM of psychotropic drugs in seminal fluid. The results showed that seminal levels of certain psychotropic drugs were detectable and correlated with alterations in sperm concentration and motility, especially in patients treated with antipsychotics. Further studies are needed to validate these findings and explore the potential impact of semen TDM in managing male fertility issues related to psychotropic medications.

Further research should focus on identifying common molecular mechanisms underlying this co-occurrence and promoting the research and treatment of developmental disorders related to the cardiovascular and central nervous systems.
