Endothelial Dysfunction-related Neurological Bleeds with Continuous Flow-Left Ventricular Assist Devices Measured by Digital Thermal Monitor

Areeba Ali, Cesar Uribe, Raquel Araujo-Gutierrez, Ana S. Cruz-Solbes, Hernan G. Marcos-Abdala, Keith A. Youker, Ashrith Guha, Guillermo Torre-Amione, Duc T. Nguyen, Edward A. Graviss, John P. Cooke, Arvind Bhimaraj

Summary: Endothelial vasodilatory dysfunction measured by a simple test utilizing digital thermal monitoring can predict adverse bleeding events in patients with CF-LVADs. Patients with poor endothelial function may have worse survival outcomes.

Haploinsufficiency of Tmem43 in cardiac myocytes activates the DNA damage response pathway leading to a late-onset senescence-associated pro-fibrotic cardiomyopathy

Leila Rouhi, Sirisha M. Cheedipudi, Suet Nee Chen, Siyang Fan, Raffaella Lombardi, Xiaofan Chen, Cristian Coarfa, Matthew J. Robertson, Priyatansh Gurha, Ali J. Marian

Summary: The study identified that TMEM43 haploinsufficiency leads to activation of the DDR and TP53 pathways, resulting in increased expression of SASP and an age-dependent pro-fibrotic cardiomyopathy. The shared mechanism in the pathogenesis of cardiomyopathies caused by mutations involving nuclear envelope proteins may involve DNA damage response.

Management of primary cardiac paraganglioma

Edward Y. Chan, Areeba Ali, Juan P. Umana, Duc T Nguyen, Dale J. Hamilton, Edward A. Graviss, Vinod Ravi, Thomas E. MacGillivray, Michael J. Reardon

Summary: Cardiac paraganglioma is a rare tumor that requires multidisciplinary management. Surgical resection yields acceptable mortality and survival rates, but complex procedures may be necessary for complete removal of the tumor and reconstruction.

Mitochondria and chronic effects of cancer therapeutics: The clinical implications

Abishai Dominic, Dale Hamilton, Jun-ichi Abe

Summary: Chemotherapy and radiation induce cellular senescence, playing crucial roles in age-related pathology. A new concept highlights the re-entry into cell cycles, rapid proliferation, and acquisition of stemness status by subsets of senescent cells induced by chemotherapy. Treatments trigger toxicity effects through damaging mitochondria, upregulating mtROS production and leading to DNA damage responses.

The Role of Calcium Signaling in Melanoma

Haoran Zhang, Zhe Chen, Aijun Zhang, Anisha A. Gupte, Dale J. Hamilton

Summary: Calcium signaling plays crucial roles in the development and progression of cutaneous melanoma. It affects melanogenesis, the tumor microenvironment, and can be targeted for therapeutic intervention.

Dynamic Tumor-Specific MHC-II Immuno-PET Predicts the Efficacy of Checkpoint Inhibitor Immunotherapy in Melanoma

Zhen Yang, Feng Li, Yuqian Huang, Na Yin, Junjun Chu, Ying Ma, Roderic I. Pettigrew, Dale J. Hamilton, Diego R. Martin, Zheng Li

Summary: Immunotherapy can benefit only a small number of patients, making the development of immuno-PET imaging crucial for predicting tumor response to immunotherapy. This study investigated the potential of tsMHC-II protein expression and its changes during interferon gamma (IFN gamma) treatment as a target for immuno-PET to predict response to immunotherapy. The results showed that tsMHC-II immuno-PET can effectively stratify tumor immunotherapy responders from nonresponders, and monitoring tsMHC-II sensitivity to IFN gamma stimulation may provide an effective strategy for predicting tumor response to immunotherapy.

Enhanced Succinate Oxidation with Mitochondrial Complex II Reactive Oxygen Species Generation in Human Prostate Cancer

Aijun Zhang, Anisha A. Gupte, Somik Chatterjee, Shumin Li, Alberto G. Ayala, Brian J. Miles, Dale J. Hamilton

Summary: The transformation of prostatic epithelial cells to prostate cancer is associated with metabolic changes and enhanced mitochondrial respiratory flux. Declining mitochondrial zinc concentrations are believed to be the molecular mechanism for this transformation. Indirect evidence based on pre-clinical models, cell lines, and tissue samples has supported the unique metabolic properties of prostate cancer cells, but direct respiratory analysis on fresh human tissue remains difficult.

Suppression of FOXP3 expression by the AP-1 family transcription factor BATF3 requires partnering with IRF4

Preston R. Arnold, Mou Wen, Lei Zhang, Yuanlin Ying, Xiang Xiao, Xiufeng Chu, Guangchuan Wang, Xiaolong Zhang, Zhuyun Mao, Aijun Zhang, Dale J. Hamilton, Wenhao Chen, Xian C. Li

Summary: This study investigated the mechanisms of IRF4 and BATF3, two transcription factors, in the conversion of CD4+ T cells to iTregs in vitro. The findings reveal that IRF4 must cooperate with BATF3 to repress FOXP3 expression and iTreg induction, and the interactions of these transcription factors antagonize glycolytic reprogramming of activated T cells, thus affecting the expression and stability of FOXP3.

The efficacy and safety of burosumab in two patients with cutaneous skeletal hypophosphatemia syndrome

Jeffrey Sugarman, Ann Maruri, Dale J. Hamilton, Laila Tabatabai, Diana Luca, Tricia Cimms, Stan Krolczyk, Mary Scott Roberts, Thomas O. Carpenter

Summary: Cutaneous skeletal hypophosphatemia syndrome is a rare disorder characterized by skeletal dysplasia and FGF23-mediated hypophosphatemia. Treatment options currently include oral phosphorus and active vitamin D analogs. However, recent studies suggest that burosumab, a fully human monoclonal antibody against FGF23, shows promising results in improving disease outcomes.

AIBP Regulates Metabolism of Ketone and Lipids but Not Mitochondrial Respiration

Jun-Dae Kim, Teng Zhou, Aijun Zhang, Shumin Li, Anisha A. Gupte, Dale J. Hamilton, Longhou Fang

Summary: This study reveals a new role of NAXE in cardiac ketone and lipid metabolism. NAXE knockout increases ketone body levels and lipid-associated metabolites while reducing cardiac lactic acid. However, it has no effect on other NAD(P)H-regulated metabolites.
