Summary: Evidence implicating p38? and p38d in inflammation is mainly based on experiments using Mapk12/Mapk13-deficient mice. A new mouse model, p38?/dKIKO, with specific depletion of p38? and p38d was generated to study their functions. The results showed reduced inflammation, susceptibility to septic shock, and Candida albicans infection in p38?/dKIKO mice compared to wild-type mice. Gene expression and phosphorylation analyses revealed the involvement of p38?/p38d in regulating innate immune responses through MEF2D phosphorylation and transcriptional activity.

Summary: Phenethyl-based edelfosine-analogs with different alkoxy substituents on the phenyl ring were designed as antitumor lipids modulating p38 MAPK activity. Saturated and monounsaturated alkoxy-substituted derivatives showed the highest antitumor activity against various cancer cells. Ortho-substituted compounds were more active than meta- or ortho-substituted compounds. Compound 1b emerged as the most potent inhibitor of p38 MAPK activity. In silico study suggested that compounds 1b and 1a could potentially bind to the lipid binding pocket of p38 MAPK. Overall, compounds 1b and 1a have broad spectrum antitumor potential through p38 MAPK modulation.

Summary: Protein tyrosine phosphatases (PTPs) have been found to play a critical role in malignant transformation and metastasis. In this study, the expression and clinical significance of Slingshot homolog 1 (SSH1), a member of PTPs, were assessed in intrahepatic cholangiocarcinoma (iCCA) patients. The results revealed that elevated SSH1 expression was associated with worse overall survival and promoted iCCA progression through the p38 MAPK-CXCL8 axis.
