Monomeric hIAPP significantly destabilizes both model and live cell membranes by increasing membrane fluidity. This interaction with membranes happens via carpet formation followed by lipid extraction in a concentration dependent manner and thus we propose that hIAPP aggregation prior to membrane interaction may not be necessary for its cytotoxicity.

Cytoskeleton-dependent clustering of membrane-bound prion protein on the cell surface

Stefanie Hackl, Xue Wen Ng, Danqin Lu, Thorsten Wohland, Christian F. W. Becker

Summary: This study focuses on the mechanism of PrPC conversion to PrPSc, revealing that PrP-GPI exhibits specific membrane accumulation and mobility, and suggests that inhibition of actin polymerization can prevent the diffusion of PrP-GPI, thus blocking the conversion of PrPC to PrPSc.

A gel-like condensation of Cidec generates lipid-permeable plates for lipid droplet fusion

Xuchao Lyu, Jia Wang, Jianqin Wang, Ye-Sheng Yin, Yun Zhu, Lin-Lin Li, Shuangru Huang, Shuang Peng, Boxin Xue, Rongyu Liao, Shi-Qiang Wang, Mian Long, Thorsten Wohland, Boon Tin Chua, Yujie Sun, Pilong Li, Xiao-Wei Chen, Li Xu, Feng-Jung Chen, Peng Li

Summary: Cidec forms a gel-like condensation at LD-LD contact sites through phase separation, promoting lipid droplet fusion by generating lipid-permeable fusion plates that are geometrically constrained by donor LDs. These Cidec condensates are unevenly distributed on the fusion plate, forming stochastic sub-compartments representing unique lipid passageways during LD fusion.

Heptanol-mediated phase separation determines phase preference of molecules in live cell membranes

Anjali Gupta, Danqin Lu, Harikrushnan Balasubramanian, Zhang Chi, Thorsten Wohland

Summary: The localization of membrane proteins within microdomains containing cholesterol and sphingolipids is crucial for cell signaling and function. However, these membrane domains are too small and dynamic to be observed with current super-resolution techniques. This study presents a simple approach to identify the phase preference of membrane-associated molecules in individual live cells within a short time. The method involves using a membrane fluidizer to induce clustering and stabilize phase separation, allowing the identification of associated molecules using routine microscopy.

The dependence of EGFR oligomerization on environment and structure: A camera-based N&B study

Harikrushnan Balasubramanian, Jagadish Sankaran, Shambhavi Pandey, Corinna Jie Hui Goh, Thorsten Wohland

Summary: This study investigates the oligomerization state of EGFR in cells and the factors influencing it. The results show that cholesterol, sphingolipids, and GM3-EGFR interactions can promote EGFR oligomerization, while the cytoskeleton does not have a significant effect. Moreover, mutations in the kinase and juxtamembrane domains of EGFR lead to a decrease in oligomerization, and EGFR phosphorylation is dependent on oligomerization.

Baird, Ulrich B. Wiesner

Summary: The study examines the interactions of fluorescent and poly(ethylene glycol)-coated (PEGylated) core-shell silica nanoparticles with biological barriers during oral delivery. It finds that ultra-small nanoparticles below 10 nm diameter passively pass through tight junctions, while particles above 20 nm diameter are actively transported through cells. Additionally, the in vivo experiments in mice demonstrate successful passage through the body and renal clearance without protein corona formation, suggesting the nanoparticles as viable candidates for oral administration.

Summary: The self-adaptability of galvanostatic electrolysis was found to be helpful in a multistage chemo- and diastereoselective electrochemically promoted cyclodimerization of chalcones. Through a series of reductive events, densely functionalized cyclopentanes with five contiguous stereocenters were obtained (25 examples, yields up to 95%, dr values up to >20:1). Further experimental and electrochemical investigations indicated the crucial role of dynamic kinetic resolution of the aldol intermediate in the reaction mechanism.
