Summary: We have conducted the first study on the use of platinum complexes in the hydrostannylation of terminal alkynes. Among several platinum complexes screened, PtCl2/XPhos exhibited the highest selectivity for beta-(E)-vinyl stannane. The catalyst system not only provided superior selectivity compared to palladium catalysis but also allowed for a telescoped hydrometallation/cross-coupling sequence, eliminating the need for excessive manipulation or purification of the intermediate stannane.

Summary: An electrophile-induced semipinacol rearrangement of cyclopropenylcarbinols is described, enabling the synthesis of polyfunctionalized cyclopropanes under mild metal-free conditions. The reaction demonstrates broad scope, accommodating various electrophiles, migrating groups, and substitution patterns on the cyclopropene. It is particularly useful for the synthesis of small ring-containing spirocycles, which are important building blocks in medicinal chemistry.

Summary: An efficient method for photocatalytic perfluoroalkylation of vinyl-substituted all-carbon quaternary centers involving 1,2-aryl migration has been developed, generating valuable multisubstituted perfluoroalkylated compounds in a single step. Mechanistically, photoinduced alkyl radical addition to an alkene leads to migration of a vicinal aryl substituent from its adjacent all-carbon quaternary center, followed by generation of a C-radical bearing two electron-withdrawing groups and reduction by a hydrogen donor to complete the domino sequence.

Summary: A novel synthetic method for polysubstituted pyrroles from isocyanides and alpha,beta-unsaturated ketones using a rhodium catalyst and bis(pinacolato)diboron (B2pin2) is described. The selectivity for different pyrrole products can be controlled by adjusting the reaction temperature and isocyanide concentration. The 2-imidoylpyrroles obtained can be hydrolyzed to isolate 2-formylpyrroles, which are naturally abundant and useful in synthesis. Deuterium-labelling experiments suggest that rhodium hydride intermediates are involved in the catalytic cycle.

Summary: A facile and efficient method for synthesizing highly substituted 2-aminopyridines has been developed in this study. The method involves in situ transformation of unstable vinyl carbodiimides using a one-pot approach. Novel highly substituted 3-functionalized 2-aminopyridines were obtained in good yields. Reaction mechanism studies suggested that Rh and potassium carbonate played crucial roles in the cyclization step.

Summary: We have developed a catalyst-controlled regiodivergent method for synthesizing structurally diverse 1,2,3,4-tetrasubstituted conjugated dienes. This method uses readily available propargyl esters and diaryliodonium salts and achieves excellent regio- and stereochemical outcomes. The unique feature of this method is the use of an in situ generated highly electrophilic aryl-Cu-III complex as both a π-Lewis acid catalyst for alkyne activation/acyloxy migration and an aryl electrophile equivalent. The competitive arylative 1,2- and 1,3-acyloxy migration patterns are controlled by Cu and Au/Cu relay catalyses, enabling the synthesis of traditionally inaccessible tetrasubstituted 1,3-dienes in a regiodivergent manner. The versatility of this method is demonstrated by its further application in the synthesis of a range of useful compounds.

Summary: Radical-based functionalization-oriented construction (FoC) is a powerful method for the direct synthesis of imine-containing azaarene derivatives. In this study, a novel FoC method was developed for the direct synthesis of highly functionalized pyrroles. This method involves the reaction of α-amino acids with easily accessible α/β-azaarene-substituted enones under visible light irradiation and with the presence of p-toluenesulfonic acid as an additive.

Summary: This study presents the design and synthesis of sulfated disaccharide inhibitors based on IdoA(2S)-GlcNS(6S), which showed potent inhibition of Sulf-2. The results suggest that IdoA(2S)-GlcNS(6S) is the shortest fragment size required for effective inhibition of Sulfs, and a trisulfated disaccharide is identified as the minimal fragment size of heparan sulfate for effective endosulfatase inhibition.

Summary: The self-adaptability of galvanostatic electrolysis was found to be helpful in a multistage chemo- and diastereoselective electrochemically promoted cyclodimerization of chalcones. Through a series of reductive events, densely functionalized cyclopentanes with five contiguous stereocenters were obtained (25 examples, yields up to 95%, dr values up to >20:1). Further experimental and electrochemical investigations indicated the crucial role of dynamic kinetic resolution of the aldol intermediate in the reaction mechanism.

Summary: In this study, the synthesis of functionalized benzo[a]phenazines and indazolo[2,3-a]quinolines was achieved through Rh(III)-catalyzed oxidative annulation of 2-arylquinoxalines and 2-aryl-2H-indazoles with allyl alcohols. The method showed a wide substrate scope, excellent functional group tolerance, good to high yields of annulated products, and the capability for scaled-up synthesis. A tentative mechanism of the annulation reaction was proposed based on a preliminary mechanistic investigation.
