This study aims to synthesize and evaluate a new generation mucoadhesive polymer, 6-maleimidohexanoic acid-grafted chitosan; MHA-CS, for transmucosal drug delivery compared with a well-known mucoadhesive polymer, Cys-CS. The successful synthesis was confirmed by nuclear magnetic resonance (NMR) spectroscopy and Fourier transform infrared (FT-IR) spectroscopy. Gel permeation chromatography (GPC) was used to determine the molecular weight of the synthesized polymer. The quantity of maleimide on the polymer chain as well as the mucoadhesive properties and biocompatibility of the synthesized polymers were also assessed. The results revealed that the synthesized MHA-CS demonstrated excellent mucoadhesive properties which is superior to CS and Cys-CS. The maleimide content bound to synthesized polymer was 466 mu mol per gram of the polymer. The cytotoxicity test assured the low toxicity of the synthesized polymer on the normal human gingival fibroblast cells. These data support the potential of MHA-CS as a novel material for developing mucoadhesive drug delivery system with enhanced mucoadhesive properties.

Summary: This study synthesized PEGDA/AA nanoparticles for the delivery of Dox to colorectal cancer cells. These nanoparticles show potential as a drug delivery carrier for targeted, staged-release therapy for colon cancer.

Topical Film-Forming Chlorhexidine Gluconate Sprays for Antiseptic Application

Summary: This study developed a topical film-forming spray for antiseptic application. By evaluating different polymers and solvents, optimal film-forming polymers and solvent systems were selected and their physicochemical properties were evaluated. The in vivo evaluation showed that the spray formed a clear and flexible film quickly with antibacterial activity. In vitro evaluations also demonstrated that the spray was non-toxic to cells and did not interfere with wound healing. Therefore, this spray could be a promising candidate for topical antiseptic application.

Chitosan capped-gold nanoparticles as skin penetration enhancer for small molecules: A study in porcine skin

Summary: The present study evaluated the effect of gold nanoparticles stabilized by chitosan (CS-AuNPs) and citrate ions (Ci-AuNPs) on skin permeation of sodium fluorescein (NaFI) and rhodamine b base (RhB). CS-AuNPs showed enhanced permeation of NaFI, superior to Ci-AuNPs, and this enhancement was likely due to improved delivery through the transepidermal pathway. However, CS-AuNPs and Ci-AuNPs had no significant effect on the permeability of the lipophilic molecule RhB. CS-AuNPs also exhibited no cytotoxicity towards human skin fibroblast cells.

Development of a novel tablet-shaped floating 3D-printed device with adjustable floating time as floating drug delivery systems provided zero-order release kinetics

Summary: This study aimed to use a 3D printer to fabricate a tablet-shaped floating device for controlling the release and floating time of domperidone tablets. The device was designed with a PVA pathway to provide a predictable floating time, and the air chamber was intentionally detached to allow elimination from the body. The optimal formulation achieved a floating time of 24 hours and a controlled drug release rate.

The dual-targeted therapy using Mcl-1 siRNA nioplex and trastuzumab showed synergistic effects in inhibiting cell growth and inducing apoptosis.

Summary: Liver disease is a major cause of global mortality, and identifying biomarkers for diagnosing its progression is crucial for improving outcomes. Targeted mass spectrometry is a powerful tool for verifying biomarker candidates and clinical applications, particularly for glycoproteins translation. However, the limitation of analyzing only one sample per run has become apparent. In this study, a high-throughput intact N-glycopeptides quantification strategy was developed, allowing the validation of 20 samples per run with an average analysis time of 3 minutes per sample. The strategy was applied in a cohort of 461 serum samples and identified a panel of 10 IgG N-glycopeptides that have strong clinical utility in evaluating the severity of liver disease.
