Summary: The expression of UBE2T was found to be significantly elevated in lung adenocarcinoma (LUAD) tissues and high UBE2T expression was associated with poor overall survival and disease-free survival. In vitro experiments showed that UBE2T modulated LUAD cell proliferation, migration, invasion, and apoptosis. Mechanistic analysis revealed that silence of UBE2T increased FBLN5 expression and inhibited the activation of p-ERK, p-GSK3 beta, and beta-catenin. In vivo experiments further confirmed that UBE2T knockdown inhibits tumor growth in LUAD.

Summary: The G protein gamma 7 (GNG7) has been found to be aberrantly expressed in certain malignant tumors, with a study focusing on its antitumor role in lung adenocarcinoma (LUAD) cells. Down-regulation of GNG7 in LUAD tissues was found to correlate with patient survival. Functional experiments demonstrated that GNG7 acts as a tumor suppressor in LUAD cells by inhibiting E2F1 and suppressing cell proliferation, migration, and invasion.

Summary: This study assessed the inhibitory effect of Ginkgolide B (GKB) on the metastatic potential of hormone receptor-positive breast cancer cells. The results showed that GKB suppressed cell migration and invasiveness by inhibiting the phosphorylation of p38 and JNK, as well as expressions and nuclear translocation of c-Jun and c-Fos. Moreover, GKB also inhibited MMP-9 activity and mRNA expressions of epithelial to mesenchymal transition markers.

These findings suggest that S. repanda could be a potential source of anti-metastasis agents in cancer therapy.

Summary: Synthetic lethality can be used to target oncogenic drivers in cancer, and in this study, it was found that WRN helicase is required for the survival of MSI CRC cells. Depletion of WRN induces p53 and PUMA, leading to apoptosis in MSI CRC cells. The vulnerability of MSI CRCs to WRN loss is mediated by p53/PUMA-dependent apoptosis. WRN depletion or treatment with ML216 suppresses the growth of MSI CRCs in vitro and in vivo in a p53/PUMA-dependent manner.

Summary: Mutant KRAS plays a crucial role in colorectal cancer by promoting Myc translation and Myc-dependent stress adaptation and proliferation. The combination of two FDA-approved drugs, Bortezomib and Everolimus, has been found to be highly effective against mutant KRAS colorectal cancer cells. This combination rapidly depletes Myc protein and induces cell death through various pathways. The study also shows that mutant KRAS colorectal cancer cells with elevated basal Myc and p-eIF2 alpha are more sensitive to the BR combination and exhibit characteristics of stress adaptation and cell death.
