Mechanistically, CTSG overexpression suppressed the Akt/mTOR signaling mechanism and elevated apoptotic-associated markers, while CTSG silencing activated the Akt/mTOR signaling mechanism and inhibited apoptotic-associated markers.

Summary: In this study, the authors investigated the role of V gamma 9V delta 2 T cells in the development and progression of psoriasis vulgaris (PV). They found that PV patients had decreased circulating V gamma 9V delta 2 T cells but enhanced proliferation and increased production of IFN-gamma and TNF-alpha by these cells. The authors also discovered that the phosphoantigen sensor butyrophilin 3A1 (BTN3A1) was expressed at higher levels on monocytes from PV patients and blocking BTN3A1 suppressed V gamma 9V delta 2 T cell activation. They further observed that CD14(+) cells in PV skin lesions highly expressed BTN3A1. These findings suggest a crucial regulatory role of BTN3A1 on monocytes in V gamma 9V delta 2 T cell activation and highlight BTN3A1 as a potential therapeutic target for psoriasis.

BCL2 inhibitors have been found to effectively kill GNA13-deficient GCB-DLBCL cells in a cell-based chemical screen. Targeting GNA13's palmitoylation enhances the sensitivity of GCB-DLBCL to BCL2 inhibitors, suggesting a potential combination therapy approach for treating GCB-DLBCL with wild-type GNA13.

Summary: MET is an oncogene that encodes a tyrosine kinase receptor for hepatocyte growth factor (HGF). When MET is mutated or amplified, it becomes a driver for cancer. The most common mutation in the MET gene affects the splicing sites of exon 14, leading to the deletion of a receptor domain. Our analysis showed that MET Delta 14 is dependent on HGF for kinase activation and has enhanced cellular protection and migration.

Summary: Prostate cancer (PCa) is a common male malignancy with unclear progression mechanisms. This study found that NRP1, highly expressed in PCa, was associated with poor prognosis in PCa patients. Functionally, NRP1 depletion inhibited PCa cell proliferation and migration, while NRP1 overexpression promoted these processes. Mechanistically, NRP1 was regulated by HIF1 alpha and interacted with EGFR, leading to EGFR phosphorylation and activation of the AKT signaling pathway, promoting PCa progression. Additionally, the NRP1 inhibitor EG01377 inactivated the EGFR/AKT signaling axis and suppressed PCa progression.

Summary: This study aimed to evaluate the expression of selected immune checkpoints in the thymus of piglets infected with two different PRRSV-1 strains. The up-regulation of PD1/PDL1, CTLA4, TIM3, LAG3 and IDO1 immune checkpoints in the thymus of PRRSV-infected piglets, especially those infected with the virulent Lena strain from 6 dpi onwards, was associated with disease progression, high viral load and cell death. Co-expression of these molecules can negatively regulate the host immune response against PRRSV by affecting T-cell development, maturation and selection.
