An Addition of U0126 Protecting Heart Grafts From Prolonged Cold Ischemia-Reperfusion Injury in Heart Transplantation: A New Preservation Strategy

Hao Zheng, Yale Su, Cuilin Zhu, Douglas Quan, Anton Skaro, Vivian McAlister, James C. Lacefield, Jifu Jiang, Peng Xue, Yefu Wang, Xiufen Zheng

Summary: Inhibition of ERK signaling with U0126 effectively attenuates prolonged cold IRI in heart transplantation. Both in vitro and in vivo experiments demonstrated the role of U0126 in reducing cell death, reactive oxygen species generation, and mitochondrial membrane potential loss. Preservation of donor hearts with U0126-supplemented solution improves graft function and reduces IRI.

Circular RNA Foxo3 in cardiac ischemia-reperfusion injury in heart transplantation: A new regulator and target

Yale Su, Cuilin Zhu, Bowen Wang, Hao Zheng, Vivian McAlister, James C. Lacefield, Douglas Quan, Tina Mele, Adam Greasley, Kexiang Liu, Xiufen Zheng

Summary: circFoxo3 plays a significant role in I/R injury during heart transplantation, and knockdown of circFoxo3 can reduce the damage and improve heart graft function through interaction with Foxo3 proteins. CircRNA is a new type of molecular regulator and a potential target for preventing I/R injury in heart transplantation.

Autoantibodies to LG3 are associated with poor long-term survival after liver retransplantation

Qingyong Xu, Vivian C. McAlister, Andrew A. House, Michele Molinari, Steve Leckie, Adriana Zeevi

Summary: Autoantibodies, such as LG3-A and AT1R-AA, are associated with poor outcomes after liver retransplantation. Patients who are double-positive for LG3-A and DSA or LG3-A and AT1R-AA have synergistic deleterious effects on regraft survival. Screening for these antibodies can help identify patients at higher risk for liver transplantation.

Chronic stress physically spares but functionally impairs innate-like invariant T cells

Patrick T. Rudak, Joshua Choi, Katie M. Parkins, Kelly L. Summers, Dwayne N. Jackson, Paula J. Foster, Anton Skaro, Ken Leslie, Vivian C. McAlister, Vijay K. Kuchroo, Wataru Inoue, Olivier Lantz, S. M. Mansour Haeryfar

Summary: Long-term psychological stress inhibits T helper 1 (T(H)1) and T(H)2-type responses by invariant natural killer T (iNKT) cells, without causing cell death. Stress triggers a split inflammatory signature in activated iNKT cells, mediated by glucocorticoid receptor signaling, and impairs their ability to enhance cytotoxicity against lymphoma or reduce metastatic melanoma burden. Stress also affects mucosa-associated invariant T (MAIT) cells' responses, but can be corrected with habituation to predictable stressors.

Managing junctional haemorrhage in the combat environment

Shane A. Smith, V. C. McAlister, L. Dubois, A. Beckett, R. Hilsden

Summary: Tactical combat casualty care and the use of extremity tourniquets have been effective in saving lives during combat, but junctional injuries present a challenge. Junctional tourniquets have emerged as a potential solution, providing a supplementary measure for persistent bleeding or hypotension in combat casualties. Surgeons need protocols for handling patients with junctional tourniquets, and evidence-based algorithms support their implementation as part of tactical combat casualty care.
