Live bacterial vaccine vectors: An overview

Genetically attenuated microorganisms, pathogens, and some commensal bacteria can be engineered to deliver recombinant heterologous antigens to stimulate the host immune system, while still offering good levels of safety. A key feature of these live vectors is their capacity to stimulate mucosal as well as humoral and/or cellular systemic immunity. This enables the use of different forms of vaccination to prevent pathogen colonization of mucosal tissues, the front door for many infectious agents. Furthermore, delivery of DNA vaccines and immune system stimulatory molecules, such as cytokines, can be achieved using these special carriers, whose adjuvant properties and, sometimes, invasive capacities enhance the immune response. More recently, the unique features and versatility of these vectors have also been exploited to develop anti-cancer vaccines, where tumor-associated antigens, cytokines, and DNA or RNA molecules are delivered. Different strategies and genetic tools are constantly being developed, increasing the antigenic potential of agents delivered by these systems, opening fresh perspectives for the deployment of vehicles for new purposes. Here we summarize the main characteristics of the different types of live bacterial vectors and discuss new applications of these delivery systems in the field of vaccinology.

Summary: In this study, a recombinant beta-xylosidase from Bacillus subtilis was immobilized on agarose and chitosan, showing high thermal stability and improved enzyme kinetic behavior. The immobilized enzymes remained efficient in corncob xylan hydrolysis after multiple cycles, indicating their potential as biocatalysts in a biorefinery context.

Summary: Mass spectrometry is crucial in proteomics, and database searching plays a significant role in protein identification. While many authors suggest teaching mass spectrometry to undergraduate students, the importance of database searching is often overlooked. Therefore, our proposal for a class specifically focusing on database searching of mass spectrometry data aims to improve students' understanding of protein identification by mass spectrometry.

Machine learning applied for metabolic flux-based control of micro-aerated fermentations in bioreactors

Summary: A machine learning control strategy, based on an artificial neural network, was proposed to enhance the productivity and yield of micro-aerobic fermentation by controlling oxygen uptake rate. Compared to traditional fermentation methods and other micro-aeration strategies, this approach demonstrated superior performance in experimental settings, improving ethanol productivity and yield.

Metabolic engineering of E. coli for pyocyanin production

Summary: This study engineered the pyocyanin biosynthetic pathway for high production in a heterologous host for the first time. Optimized culture and induction parameters led to a 3.5-fold increase in pyocyanin accumulation, while pathway balancing through testing plasmids with different copy numbers resulted in a fourfold difference in product titer among the engineered strains. Co-expression of Vitreoscilla hemoglobin Vhb further improved pyocyanin production to a final titer of 18.8 mg/L, demonstrating the potential of using Escherichia coli for phenazines production in electro-fermentation systems.

Summary: A strategy based on strain evaluation and integration with 1G ethanol production was used to develop a robust and scalable process for 2G ethanol production. The study found that using mixed medium of crude bagasse hydrolysate and molasses achieved high ethanol concentrations and long-term stability.

Summary: Citrus canker is a bacterial disease that affects citrus crops worldwide. This study reports the isolation and characterization of a new jumbo phage, LucasX, which can infect Xanthomonas citri and X. fuscans. LucasX has a large genome and shows potential as a biocontrol agent for citrus canker.

Summary: This study successfully achieved the separation of FVIII and PCC by directly loading plasma onto anion exchange resin under high ionic strength. Using certain resins, blood products can be purified quickly without the need for cryoprecipitation.

Teaching cellular metabolism using metabolic model simulations

Summary: This article introduces the use of simulation tool OptFlux to teach cellular metabolism to chemical engineers. Students explore the quantitative aspects of cellular metabolism by simulating the behavior of Escherichia coli and Saccharomyces cerevisiae cells under different environmental conditions. The students' feedback regarding the use of OptFlux as an educational tool is highly positive, with most students finding it a useful teaching strategy.
