The role of apoptosis in the ageing process is still unclear, but this study shows that Fas signaling can induce premature senescence in mouse embryonic fibroblasts, while Fas deficiency leads to senescence through increased SA-beta-galactosidase activity, p53 phosphorylation, and the expression of p16, p21, and IL-6.

Summary: This study investigates the mechanisms underlying the effects of iron on the barrier function of human endothelium. The experimental results suggest that exposure to iron leads to endothelial cell elongation, upregulation of stress-induced proteins, and a dose-dependent drop in endothelial integrity. Reactive oxygen species (ROS) production and the role of low-grade inflammation were also investigated. The findings indicate that iron-induced damage to the vascular endothelium could contribute to increased cardiovascular risk.

Summary: Inhibition of Thioredoxin Reductase (TrxR) has been identified as a promising strategy to disrupt AML metabolism, as it impairs leukemic cell growth and leads to cell death. TrxR is a major regulator of metabolism in AML cells, impacting glycolysis and the pentose phosphate pathway (PPP). The combined inhibition of TrxR and PPP enhances leukemia growth inhibition, highlighting the potential of TrxR abrogation, particularly with S-250, as an effective therapeutic approach for AML.
