Comparison of various microbial inocula for the efficient anaerobic digestion of Laminaria hyperborea

Background: The hydrolysis of seaweed polysaccharides is the rate limiting step in anaerobic digestion (AD) of seaweeds. Seven different microbial inocula and a mixture of these (inoculum 8) were therefore compared in triplicate, each grown over four weeks in static culture for the ability to degrade Laminaria hyperborea seaweed and produce methane through AD. Results: All the inocula could degrade L. hyperborea and produce methane to some extent. However, an inoculum of slurry from a human sewage anaerobic digester, one of rumen contents from seaweed-eating North Ronaldsay sheep and inoculum 8 used most seaweed volatile solids (VS) (means ranged between 59 and 68% used), suggesting that these each had efficient seaweed polysaccharide digesting bacteria. The human sewage inoculum, an inoculum of anaerobic marine mud mixed with rotting seaweed and inoculum 8 all developed to give higher volumes of methane (means between 41 and 62.5 ml g(-1) of seaweed VS by week four), compared to other inocula (means between 3.5 and 27.5 ml g(-1) VS). Inoculum 8 also gave the highest acetate production (6.5 mmol g(-1) VS) in a single-stage fermenter AD system and produced most methane (8.4 mL mmol acetate(-1)) in phase II of a two-stage AD system. Conclusions: Overall inoculum 8 was found to be the most efficient inoculum for AD of seaweed. The study therefore showed that selection and inclusion of efficient polysaccharide hydrolysing bacteria and methanogenic archaea in an inoculum offer increased methane productivity in AD of L. hyperborea. This inoculum will now being tested in larger scale (10L) continuously stirred reactors optimised for feed rate and retention time to determine maximum methane production under single-stage and two-stage AD systems.

Summary: This study investigates the plasticity of the polar lipid profile of marine and freshwater Nannochloropsis species grown in indoor and outdoor photobioreactors. The results show that indoor conditions lead to higher lipid content, while outdoor conditions result in a lipid profile rich in phospholipids and betaine lipids. The study provides insight into the lipid metabolic adaptation of Nannochloropsis species.

Optimisation of Biomass Production and Nutritional Value of Two Marine Diatoms (Bacillariophyceae), Skeletonema costatum and Chaetoceros calcitrans

Summary: The aim of this study was to improve the biomass productivity of two high-value diatom species by optimizing the culture medium. The results showed significant increases in biomass productivity and the content of omega-3 fatty acids for both diatoms. This research provides the basic tools for the industrial cultivation of these diatoms, enhancing aquaculture and nutraceutical production.

Biogas Production from Microalgal Biomass Produced in the Tertiary Treatment of Urban Wastewater: Assessment of Seasonal Variations

Summary: The valorization of microalgal biomass during wastewater treatment can mitigate treatment costs and create an energy surplus. In this study, biomass obtained from pilot photobioreactors for tertiary wastewater treatment was used to produce biogas. The results showed that the implementation of this system could reduce energy consumption by 20% and contribute towards meeting the EU's decarbonization targets.

Structural characterization of exopolysaccharides obtained from Porphyridium cruentum exhausted culture medium

Summary: The study aimed to recover extracellular polysaccharides (EPS) from the exhausted culture medium (ECM) of Porphyridium cruentum using different extracting and purifying methods for future biotechnological applications. The purification methods, including precipitation with cold absolute ethanol, ultrasound, dialysis, and trichloroacetic acid (TCA), resulted in lower yield but higher carbohydrate content. TCA purification provided twice as much total carbohydrates as the non-purified samples, mainly composed of xylose, galactose, and glucose. Scanning Electron Microscopy (SEM) images revealed the smooth structure of P. cruentum EPS, with TCA post-treatment and dialysis yielding larger and purest particles suitable for film making. Microalgal polymeric by-products are a sustainable source of valuable compounds, and purification treatment is an important step in valorizing this material.

Summary: The aim of this study was to evaluate the efficiency of the new GreenDune photobioreactors in tertiary wastewater treatment, treated wastewater reuse, and biomass application. The results showed that the system was effective in removing the main pollutant in wastewater and the produced biomass had potential for biofuel production and use as biofertilizers.

Summary: In recent years, the large-scale cultivation and commercialization of microalgae/cyanobacteria biomasses have become a worldwide trend, mainly directed to the animal and human nutrition markets due to their outstanding nutritional value. Crocosphaera chwakensis CCY0110 is a marine unicellular cyanobacterium that produces a promising and versatile extracellular carbohydrate polymer (Cyanoflan). Therefore, envisaging a biorefinery approach with a multi-product stream (zero-waste), C. chwakensis biomass was cultivated at pilot-scale and its nutritional composition was evaluated.

Dunaliella viridis TAV01: A Halotolerant, Protein-Rich Microalga from the Algarve Coast

Summary: Dunaliella viridis TAV01 is a new strain with beneficial features including tolerance to harsh environmental conditions, high growth rates, and amino acid composition suitable for human consumption. Its identification was confirmed at the species level through evolutionary analysis of ITS1 and ITS2 regions, and supported by secondary structure analysis. TAV01 exhibits high protein and lipid contents, predominantly polyunsaturated fatty acids, and a balanced essential amino acid profile. The major carotenoid present is lutein. These qualities make D. viridis TAV01 a potential valuable resource for novel feed or food products.
