The heterogeneity of epilepsy syndromes and pathologies creates a great challenge for the search for biomarkers. Not surprisingly, identification of a marker that is specific and sensitive for a given epileptogenic pathology remains an unmet need. There have, however, been several studies of major epileptogenic etiologies like traumatic brain injury that aimed to identify molecular markers in blood and cerebrospinal fluid that predict outcome, by using proteomics and metabolomics. Unfortunately, epileptogenesis has not been analyzed as an outcome measure. Another question to be explored is whether a palette of molecular markers is needed, rather than a single molecule, with each marker probing a different component of epileptogenic pathology. Further, perhaps multiple biomarker platforms (e.g., imaging, proteomics, electrophysiology) should be used in combination and/or in a defined temporal sequence.

Summary: The study analyzed behavior in a rat model of epilepsy, identifying differences in behavioral impairments between animals with short versus long latency to seizures and low versus high number of seizures. Results suggest that epilepsy intensity and seizure frequency may affect animal behavior, with mild behavioral changes observed overall.

Summary: The study identified a 592 bp rat Ttyh1 promoter sequence and found that Ascl1, Sp1, and responsive cis-regulatory elements may be involved in the transcriptional regulation of Ttyh1 expression in rat dissociated hippocampal neurons in vitro, providing novel information about Ttyh1 gene regulation in neurons.

Emerging Molecular Targets for Anti-Epileptogenic and Epilepsy Modifying Drugs

Summary: The pharmacological treatment of epilepsy is only aimed at relieving symptoms. Despite years of research, a causal treatment for this common neurological disorder is still unavailable. Advances in neuroscience and molecular biology tools, as well as improved animal models, may accelerate the development of antiepileptogenic and epilepsy-modifying drugs. Epileptogenesis triggers various changes in the genome, epigenome, and transcriptome, leading to the transformation of neuronal circuits and the occurrence of seizures.
