Summary: In this study, a potential compound was identified that could attenuate the progression of osteoarthritis by inhibiting matrix-degrading enzymes. The compound showed a dose-dependent decrease in mRNA expression of MMP13 and ADAMTS9 and had a synergistic effect when combined with low-dose betamethasone.

A newly designed inhibitor, named inhibitor 3, with improved solubility and stability, effectively inhibited MMP-13-mediated osteoclastogenesis and is considered a potential next-generation probe for breast cancer metastasis in vivo studies.

Summary: This study demonstrated that 30 kDa FN-f can increase pro-inflammatory gene expression in articular chondrocytes, meniscal fibrochondrocytes, and synovial fibroblasts, which was significantly attenuated by PRP treatment. PRP treatment also attenuated MMP-13 expression induced by 30 kDa FN-f through the TLR2 signaling pathway.

Hydroxamate and carboxylic acid compounds were found to be effective inhibitors, and solvent accessibility, hydrogen bond donor and acceptor groups, and bulky aromatic groups were found to be important for interactions at the MMP-2 active site.

Summary: Identifying serum biomarkers associated with rheumatoid arthritis (RA) activity is important for understanding disease progression, prognosis, and quality of life. This study analyzed the presence and clinical use of MMP-13, VEGF, TNF-a, and IL-6 in RA patients and found higher levels compared to controls. Correlations were identified between MMP-13, VEGF, disease activity, and ultrasound findings. MMP-13 and VEGF can be used as biomarkers to provide a better understanding of inflammation, disease activity, and damage in RA patients.

Summary: Fibrostenotic disease is a common complication in Crohn's disease characterized by ECM accumulation. This study found that MMP13 expression is regulated by IL36R signaling during intestinal fibrosis, and the loss of MMP13 can alleviate intestinal fibrosis in mice. Targeting IL36R-inducible MMP13 could be a promising approach to interfere with the development and progression of intestinal fibrosis.
