Virulence domain of the RYMV genome-linked viral protein VPg towards rice rymv1-2-mediated resistance

Virulent variants of Rice yellow mottle virus (genus Sobemovirus) can emerge on the highly resistant rice cultivars Gigante and Bekarosaka. Non-synonymous mutations responsible for the breakdown of the recessive resistance gene rymv1-2 were located in the VPg after determination of its termini in the polyprotein P2a. The secondary structure of this protein was predicted to include a central alpha-helix. The two major amino acids related to virulence are located on the same side of this helix. The 3D topology and the biochemical properties of virulence mutations both suggested a direct site-to-site interaction between RYMV VPg and rice eIF(iso)4G encoded by rymv1.

Summary: This study investigated the genetic diversity of the rice stripe necrosis virus (RSNV) and found unexpected differences with the beet necrotic yellow vein virus (BNYVV), suggesting a complex evolutionary history of the genus Benyvirus. Summary: Pearl millet is a staple food widely cultivated in sub-Saharan Africa, and a new virus called Pennisetum glaucum marafivirus was discovered in Burkina Faso in 2018, belonging to the Marafivirus genus. The complete nucleotide sequence of PGMV shares 68.5% identity with sorghum bicolor marafivirus and its coat protein shares 58.5% identity with oat blue dwarf virus. Phylogenetic analysis confirms that the pearl millet virus is grouped with Marafivirus genus members in the Tymoviridae family.

Summary: The RYMV3 gene in rice codes for a NLR immune receptor that detects the rice yellow mottle virus (RYMV) by forming a recognition complex with the viral coat protein (CP). The virus escapes detection by mutating its CP, interfering with the formation of the recognition complex. This study reveals the immune function of NLRs in monocotyledonous plants and provides insights into virus adaptation and the development of resistance against viral diseases.
