Inhibitory effects of (-)-loliolide on cellular senescence in human dermal fibroblasts

Cellular senescence influences tumor suppression and progress, tissue repair and regeneration, tissue and organismal aging, and age-related diseases. Aging intervention might be an advantageous target for prevention and treatment of diverse age-related diseases. In this study, we investigated whether (-)-loliolide purified from the crude extract of Polygonum aviculare exerted inhibitory activity against cellular senescence in human dermal fibroblasts (HDFs). (-)-Loliolide diminished senescence-associated beta-galactosidase activity (SA-beta-gal), the level of p21 protein, and the level of reactive oxygen species in senescent cells induced by adriamycin treatment. (-)-Loliolide also attenuated SA-beta-gal activity in HDFs under replicative senescence. These findings imply that (-)-loliolide rescues cellular senescence in HDFs and might be useful for the development of dietary supplements or cosmetics that ameliorate tissue aging or age-associated diseases.

Summary: Recent studies have shown that a single dose of mFK+CLO can induce tolerance to islet xenografts by reducing the antigen presenting and T cell activation capacity of dendritic cells, as well as promoting the generation of regulatory T cells. This immunosuppressive protocol may pave the way for improving therapeutic outcomes of clinical islet transplantation by enhancing the generation of tolerogenic dendritic cells and regulatory T cells.

Summary: This study found that CD9 antibody-modified nanoparticles showed high cellular uptake in vitro models, reducing oxidative stress and inflammation in senescent cells, and improving cell viability. In vivo experiments demonstrated that these nanoparticles effectively targeted senescent areas, delivering anti-senescence drugs to atherosclerotic plaques and alleviating the progression of atherosclerosis. This suggests a novel and precise therapeutic approach for treating atherosclerosis by targeting senescent cells.

Summary: This study reports the potential of novel peptides MIF1 and MIF2 derived from MSTN in enhancing myogenesis and reducing adipogenesis. Modified MIF1 and MIF2 showed better cell proliferation and differentiation compared to non-modified peptides. Pretreatment of mice with (Ac)-MIF1 or (Ac)-MIF2-(NH2) promoted muscle regeneration.
