QUERCETIN AND LITHIUM CHLORIDE MODULATE WNT SIGNALING IN PLURIPOTENT EMBRYONAL CARCINOMA NT2/D1 CELLS

Wnt signaling functions in numerous cellular activities such as cell fate determination, patterning, and migration in embryogenesis, apoptosis, etc. In this study, we used quercetin and lithium chloride to investigate modulations of the Wnt signaling pathway in human pluripotent embryonal carcinoma NT2/D1 cell line. First, we optimized conditions for NT2/D1 cell treatments with quercetin and lithium chloride and assessed their cytotoxic effects on the cells, cell viability and proliferation rate. Our results showed that induction of cell death by quercetin and LiCl is p53-dependent in NT2/D cells. We also examined the degree of Wnt signaling modulations by analyzing the expression of c-myc, a well-known Wnt signaling target gene. Since the retinoic acid induction of NT2/D1 cells is good in an in vitro model system for human neural differentiation, studying Wnt signaling modulation in NT2/D1 would contribute to a better understanding of the mechanisms involved in neural stem cell maintenance and human neural development.

Bioactivities of Salvia nemorosa L. inflorescences are influenced by the extraction solvents

Summary: The study explored the impact of different solvents on the bioactivities of Salvia nemorosa L. inflorescences, highlighting butanol as the most suitable solvent for wound healing improvement and ethanol/water combination for the highest antioxidant potential. The extracts showed potential in antimicrobial and antibiofilm activities, with one extract demonstrating immunomodulatory action and antibacterial mechanism. Overall, S. nemorosa inflorescences were found to be rich sources of bioactive polyphenols with various therapeutic applications.

Blue luminescent amino-functionalized graphene quantum dots as a responsive material for potential detection of metal ions and malathion

Summary: Non-toxic, cheap, easy, rapid, and sensitive methods are needed for the detection of large amounts of hazardous and toxic substances in the environment. Blue luminescent graphene quantum dots (GQDs) were produced by electrochemical cleavage of graphite electrodes followed by gamma irradiation in the presence of ethylenediamine (EDA), and they were able to detect metal ions. Additionally, gamma irradiated GQDs acted as an ultra-sensitive turn-on probe for Malathion detection.

Structural, optical, and bioimaging characterization of carbon quantum dots solvothermally synthesized from o-phenylenediamine

Summary: In this study, carbon quantum dots were prepared from o-phenylenediamine dissolved in toluene by a solvothermal route, and then encapsulated into polyurethane films by a swelling-encapsulation-shrink method. The precursor was found to have a significant influence on the structural, chemical, and optical properties. Antibacterial and cytotoxicity tests showed low antibacterial potential and dark cytotoxicity, while cellular uptake experiments confirmed the potential use of these dots as bioimaging probes.

Highly Efficient Antibacterial Polymer Composites Based on Hydrophobic Riboflavin Carbon Polymerized Dots

Summary: The development of new types of antimicrobial coatings is of great significance in tackling the increasing problem of pathogen transmission from infectious surfaces. This study presents highly potent antimicrobial polyurethane composite films encapsulated by hydrophobic riboflavin-based carbon polymer dots. These composites can effectively eradicate Escherichia coli and Staphylococcus aureus within a short period of time under low-power blue light, and show low toxicity to cells. They can be used for sterilization of frequently touched objects in the healthcare industry.

Crosstalk between SOX Genes and Long Non-Coding RNAs in Glioblastoma

Summary: Glioblastoma (GBM) is the most devastating primary brain malignancy, and the prognosis for most GBM patients remains poor. The interplay between SOX genes and lncRNAs plays important roles in tumor initiation, progression, therapy resistance, and recurrence in GBM. This review systematically summarizes the current literature data on the complex interplay between SOX genes and lncRNAs in GBM and highlights its significance in searching for new biomarkers and therapeutic approaches.

Carbon quantum dots, a new member of the carbon nanomaterials family, show potential as visible-light-triggered antibacterial agents. This study investigates the antibacterial and cytotoxic activities of gamma-ray-irradiated carbon quantum dots, finding that they have antibacterial activity and are non-toxic to human cells.

The Role of SOX Transcription Factors in Ageing and Age-Related Diseases

Summary: The pursuit of eternal youth and immortality dates back to ancient times. Ageing is a natural process accompanied by functional declines, which contribute to age-related diseases. Stem cell exhaustion is a hallmark of ageing, and the SOX transcription factors have significant roles in stem cell self-renewal and differentiation. Understanding the molecular mechanisms of ageing and the involvement of SOX transcription factors can help develop strategies for delaying ageing and preventing age-related diseases.

Summary: Precise tuning of gene expression, accomplished by regulatory networks of transcription factors, epigenetic modifiers, and microRNAs, is crucial for proper neural development and brain function. The complex regulatory network between SOX transcription factors and microRNAs plays important roles in neurogenesis. Deregulated SOX/microRNA interplay has been associated with various brain pathologies. Therapeutic strategies targeting the interplay have emerged as a promising tool for neural tissue regeneration and neurorestoration.

CHEMICAL ENGINEERING METHODS IN ANALYSES OF 3D CANCER CELL CULTURES: HYDRODYNAMIC AND MASS TRANSPORT CONSIDERATIONS

Summary: A multidisciplinary approach was utilized in this study to develop a biomimetic system for 3D cultures of cancer cells. Cancer cell lines were immobilized in alginate microbeads and microfibers and cultured under different conditions. Chemical engineering methods were used to explain the results obtained. The study showed that fluid shear stresses affected the viability of cancer cells, and different velocities had different effects on the proliferation of glioma cells. Moreover, the mass transport within the microfibers was described using a diffusion-advection-reaction model.

Summary: This study explores the anti-neurodegenerative and anti-glioblastoma potential of Phlomis fruticosa extract. The extract was found to reduce the activities of enzymes linked to neurodegenerative diseases and exhibit excellent antioxidant potential. It also showed cytotoxic effects on glioblastoma cells and increased the production of ROS. Twelve different polyphenols were identified in the extract, with cis 3-O-caffeoylquinic acid being the most abundant.
