Summary: The Glasgow Microenvironment Score (GMS) is validated as a prognostic tool for patients with stage I-III colorectal cancer, independent of TNM staging, and has the ability to stratify both low- and high-risk disease. Additionally, GMS 0 can be used to identify patients who benefit from FOLFOX over CAPOX chemotherapy.

Summary: In triple-negative breast cancer (TNBC) cells, DYRK2 phosphorylates HSF1 to promote its nuclear stability and transcriptional activity, with DYRK2 depletion reducing HSF1 activity. Moreover, DYRK2 levels positively correlate with active HSF1 in TNBC tumors and associate with poor prognosis, suggesting DYRK2 may be promoting TNBC.

Summary: The analysis confirmed a significant association between high expression of HIF-1 alpha and poorer disease-free survival (DFS) and overall survival (OS) in breast cancer patients. Various factors such as survival analysis, study region, antibodies used, scoring methods, and threshold methods were found to impact the expression of HIF-1 alpha.

Summary: KRAS-mutant colorectal cancers exhibit resistance to therapeutics, but targeting the mTORC1 and MNK pathways can sensitize them to rapalogs. This approach has shown effectiveness in mouse models and human organoids, offering a potential strategy for patients with elevated c-MYC expression.

Summary: This study demonstrates that the pre-operative systemic inflammatory response (SIR) in patients undergoing curative surgery for colon cancer is best evaluated using a Systemic Inflammatory Grade (SIG) combining modified Glasgow Prognostic Score (mGPS) and neutrophil-lymphocyte ratio (NLR). Both mGPS and NLR have a significant impact on survival rates in colon cancer patients, and their combined use can better predict outcomes.

Summary: Colorectal cancer is the third most common cancer worldwide. Poor survival and disease metastasis are associated with the development of tumor metastasis in CRC patients, making it important to study potential biomarkers for predicting outcomes. Tumor budding is an independent prognostic marker for poor survival and disease metastasis, and it has been suggested to be included in routine clinicopathological factors for risk assessment. However, there is currently no clear evidence regarding the underlying molecular mechanism driving this phenotype, or the relationship between tumor budding and the tumor microenvironment.

Summary: Preserving proteostasis is a crucial survival mechanism for cancer, and targeting the kinase DYRK2 and its substrate HSF1 can induce apoptosis faster than individual targeting. This study suggests that cotargeting DYRK2 and HSF1 could be a promising strategy in suppressing cancer.
