Tumor-targeting and pH-sensitive lipoprotein-mimic nanocarrier for targeted intracellular delivery of paclitaxel

In the present study, we constructed a tumor-targeting and pH-sensitive lipoprotein-mimic nanocarrier containing paclitaxel (FA-BSA-LC/DOPE-PTX), by adding 1,2-dioleoyl-sn-glycero-3-phosphoethanol-amine (DOPE) and oleic acid as pH-sensitive components into the formulation of lipid core and then coating with folic acid modified bovine serum albumin (FA-BSA) for tumor targeting activity. In vitro drug release study demonstrated that paclitaxel (PTX) was released from FA-BSA-LC/DOPE in a pH-dependent manner. The vitro cytotoxicity assays showed that all the blank nanocarriers were nontoxic. However, MTT assay showed that FA-BSA-LC/DOPE-PTX was highly cytotoxic. Cellular uptake experiments analyzed with flow cytometry and laser scan confocal microscope (LSCM) revealed that FA-BSA-LC/DOPE was taken up in great amount via folate receptor-mediated endocytosis and pH-sensitive release of drug to cytoplasm. Furthermore, the study of intracellular drug release behavior demonstrated that the FABSA-LC/DOPE escaped from lysosomes and released drug into cytoplasm. The in vivo targeting activity showed that the nanocarrier selectively targeted tumor and had long residence time for BSA layer increased the stability in blood. Moreover, FA-BSA-LC/DOPE-PTX produced very marked anti-tumor activity in tumor-bearing mice in vivo. Therefore, FA-BSA-LC/DOPE as biocompatible, tumor-targeting and pH-sensitive lipoprotein-mimic nanocarrier is a promising system for effective intracellular delivery of PTX to tumor. Reversing ferroptosis resistance by MOFs through regulation intracellular redox homeostasis

Summary: Ferroptosis is a potential strategy to overcome cancer chemotherapy resistance, but cancer cells can develop resistance to ferroptosis through antioxidative defense mechanisms. In this study, a BSO-loaded metal organic framework (BMH) was prepared to enhance oxidative damage and inhibit antioxidative defense. The results showed that BMH restored the susceptibility of cancer cells to ferroptosis under irradiation and exhibited superior anti-tumor performance in vivo.

Community-integrated multi-omics facilitates screening and isolation of the organohalide dehalogenation microorganism

Summary: This study investigated the microbial community composition and potential organohalide dehalogenation bacteria (OHDBs) in industrial saponification wastewater using 16S rRNA sequencing and metagenomic sequencing. The study found that the dominant microbial phyla in the wastewater were Proteobacteria and Patescibacteria, and identified six bacterial groups as biomarkers for organohalide removal. A total of 105 hydrolytic dehalogenase genes were also identified in 42 reconstructed metagenomes, indicating the potential for organohalide hydrolytic dehalogenation in the microbial community. Additionally, the study characterized the dehalogenation activities of an isolated OHDB, Microbacterium sp. J1-1.

Nanoparticle-inhibited neutrophil elastase prevents neutrophil extracellular trap and alleviates rheumatoid arthritis in C57BL/6 mice

Summary: Rheumatoid arthritis (RA) is characterized by inflammation outbreak and joint destruction. Neutrophils play a crucial role in RA by releasing excess neutrophil extracellular traps (NETs), but preventing NETs formation is still challenging. In this study, a method using sivelestat-conjugated bovine serum albumin nanoparticles (BTST NPs) was established to selectively inhibit neutrophil elastase (NE) and prevent NETs production. Additionally, the BTST NPs were loaded with dexamethasone palmitate (DP) for synergistic treatment of RA. The results showed that DP/BTST NPs could effectively target inflamed neutrophils, hinder NETs generation, and improve RA symptoms.

Chemoproteomic mapping of the glycolytic targetome in cancer cells

Characterization of ZmPMP3g function in drought tolerance of maize

Summary: It was found that overexpression of the ZmPMP3g gene in transgenic maize lines enhanced drought tolerance by improving root growth, increasing antioxidant capacity, maintaining membrane lipid integrity, and regulating intracellular osmotic pressure. Additionally, ZmPMP3g overexpression affected the balance of ABA-GA1-GA3, resulting in increased drought tolerance. These findings provide insights into the mechanisms underlying maize drought tolerance and suggest a potential strategy for improving crop resilience to abiotic stresses.

Spraying exogenous regulators improves breadmaking quality and modifies the spatial distribution of gluten protein

Summary: This study investigated the effects of spraying different combinations of exogenous regulators on the breadmaking quality of flour. Three spraying schemes were screened out to improve protein contents and yield in the flour. The amounts of different proteins in the flour were increased, and bread volume was also increased. The M treatment showed superior gluten quality and breadmaking quality in comparison to G and C treatments.

Copper-ion-mediated removal of nitrous oxide by a salt-tolerant aerobic denitrifier Halomonas sp. 3H

Summary: In this study, a halotolerant bacterium Halomonas sp. 3H was isolated from a eutrophic saline lake sediment, which could remove ammonium nitrogen and nitrate/nitrite nitrogen via aerobic denitrification in high-salinity wastewater and reduce the production of greenhouse gas N2O. The strain could also remove added N2O with the further addition of Cu2+, showing its potential to mitigate complex and significant climate changes.

Meta-omics assisted microbial gene and strain resources mining in contaminant environment

This review highlights the applications and methodologies of meta-omics in uncovering genes and microbes from contaminated environments, which may contribute to future bioremediation research.

Implantable metaverse with retinal prostheses and bionic vision processing

Summary: We propose an implantable metaverse with retinal prostheses that work in tandem with bionic vision processing. Our approach differs from conventional retinal prostheses by rearranging electrodes to match the distribution of ganglion cells. To emulate human vision, we develop a scheme for bionic vision processing that visualizes monocular image, binocular image fusion, and parallax-induced depth map on a three-dimensional eye model.

3D human foreskin model for testing topical formulations of sildenafil citrate

Summary: This study aimed to investigate the loading of sildenafil citrate in three commercial transdermal vehicles using 3D full-thickness skin equivalent and compare the results with permeability experiments using porcine skin. The results showed that the results obtained using the 3D skin equivalent were comparable to those obtained using porcine skin, suggesting that the 3D skin model can be a valid alternative for ex-vivo skin absorption experiments.
