Summary: Adipose tissue plays an important role in health and metabolic diseases. Deficiency of lipid kinase PIK3C3/VPS34 in adipocytes leads to defects in adipocyte differentiation, survival and function, resulting in reduced adipose tissue mass, abnormal blood lipid levels, fatty liver disease, diabetes, and impaired body temperature control. These findings suggest that targeting adipocyte PIK3C3/VPS34 could be a potential therapeutic intervention for metabolic diseases.

Summary: PIK3C3 plays a critical role in T cell metabolism and function, with its deficiency leading to impaired metabolism and differentiation of T cells, as well as reduced levels of active mitochondria. Interestingly, Pik3c3 deletion did not significantly affect the ability of animals to clear tumor metastases.

Summary: PIK3C3, as a member of the PI3K family, plays a critical role in autophagy and tumor development. Inhibition of PIK3C3 can suppress autophagy and affect tumor cell proliferation, growth, and invasion. Discovery of pharmacological inhibitors targeting PIK3C3 may provide new strategies for improving treatment outcomes in PIK3C3-mediated diseases.

Summary: The relationship between predicted functional consequences of missense mutations in genes related to lipid metabolism and lipid phenotypes is weak. Only nine out of 72 associated SNPs showed consistent predicted functional mutations and were not associated with expected lipid traits affected by gene inactivation. The study suggests that current bioinformatic tools have limited ability to predict the impact of missense mutations on complex phenotypes.

Summary: This study discovered an association between variations in the porcine CISH gene and susceptibility to diarrhea in pigs, as well as identified a novel 12-bp indel mutation. In vitro experiments further supported the functional role of this variation.

Summary: Phosphatidylinositol-3-phosphate (PtdIns3P) plays a crucial role in autophagosome formation and endocytic trafficking regulation, with the activities of human PIK3C3/VPS34-containing complexes I and II being significantly influenced by membrane physicochemical parameters such as lipid unsaturation, curvature, and negative charge. The interaction of these complexes with membranes, specifically the presence of unsaturated lipids, membrane curvature, and negatively charged phospholipids, is essential for regulating their kinase activity and ultimately their function in autophagy and endocytosis.

Summary: In this study, the researchers found a SNP in NLRP9 that is associated with litter size in Large White pigs. Sows with the CC genotype have the highest total number of piglets born per litter and number of piglets born alive per litter, while sows with the TT genotype have the lowest. There is also a significant relationship between NLRP9 polymorphism and average daily gain.

Summary: This study revealed the expression and polymorphism of GPAT3 in its promoter region in Laiwu pigs, which is consistent with changes in intramuscular fat content. The expression level of GPAT3 was found to be higher in Laiwu pigs, and a SNP at -1526 site may contribute to IMF deposition by affecting its expression in pigs.

Summary: The lipid kinase PIK3C3 plays a crucial role in maintaining adipose tissue homeostasis and systemic metabolism. Its deficiency disrupts adipocyte autophagy and hinders adipocyte differentiation, survival, and function, leading to abnormalities in adipose tissue and the development of metabolic diseases.

Summary: This study conducted molecular detection and genome sequencing of Japanese encephalitis virus (JEV) isolates from pigs and mosquitoes in Assam, India. The analysis revealed that these isolates belong to genotype III and are closely related to strains from China, indicating the transboundary circulation of JEV genotype III through mosquito-swine transmission cycles.
