Hydroxyflavones as a New Family of Matrices for MALDI Tissue Imaging

The discovery of new matrices that are suitable for in situ analysis of low molecular-weight compounds by matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS) is an important technological aspect of tissue imaging. In this work, ten natural flavonoid compounds, including flavone and nine of its mono- or polyhydroxyl-substituted analogues (3-hydroxyflavone, 5-hydroxyflavone, 3,7-dihydroxyflavone, chrysin, 7,3',4'-trihydroxyflavone, fisetin, luteolin, quercetin, and morin) were evaluated as potential MALDI matrices for the profiling and imaging of endogenous lipids in mouse liver, using a Fourier transform ion cyclotron resonance (FTICR) mass spectrometer with a 355-nm Nd:YAG UV laser, in the positive ion mode. When an electronic sprayer was used for matrix coating and with a high-pH (0.1-0.5% ammonia hydroxide) matrix solvent, eight of the ten compounds, all of which had at least one OH group at the C3 or C5 position of the flavone structure, enabled the successful detection of 77 to 161 phospholipids and other lipids. The best results were observed with two penta-OH flavones (i.e., quercertin and morin). Taking quercetin as an example, this matrix showed characteristics superior to those of commonly used MALDI matrices, such as DHB (2,5-dihydroxybenzoic acid), CHCA (alpha-cyano-4-hydroxycinnamic acid), and 2-mercaptobenzothiazole (2-MBT). These characteristics were: mu m-sized matrix crystals, uniform matrix coating, low volatility in the high vacuum (similar to 10(-7) mbar) source, good chemical stability, low yield of matrix-related ions, low matrix consumption, low power threshold for laser desorption/ionization, and improved safety of handling. The use of quercetin led to improved lipid imaging, with 212 lipids being successfully imaged from rat brain in a single experiment and with asymmetric distributions of some lipids in left and right brain hippocampus being observed for the first time.

Summary: This study demonstrated that caffeic acid (CA) can effectively enhance the detection and imaging of high-molecular-weight (HMW) proteins in biological tissue sections through matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI). Compared to sinapinic acid (SA) and ferulic acid (FA), CA showed superior performance in detecting HMW proteins in biological tissues. Additionally, the use of a CA matrix significantly improved the imaging of proteins on selected tissue sections, demonstrating its potential as a standard organic acid matrix for tissue imaging of HMW proteins.

Mass Spectrometry Imaging of Lipids in the Scent Glands of Muskrat (Ondatra zibethicus) in Different Reproductive Statuses

Summary: In this study, the distribution and expression of lipids in the scent glands of muskrats during different reproductive statuses were investigated using imaging mass spectrometry and quantitative real-time PCR. The results showed significant differences in lipid expression and spatial distribution in the scent glands of muskrats during different reproductive statuses. Additionally, the expression levels of certain lipid molecules and lipid synthesis enzymes were higher during the breeding season. The study suggests that seasonal changes in lipid metabolism may affect the function of muskrat scent glands.

Summary: This study investigates the regulatory influences of 6-BA and KT in the rapid micropropagation system of A. hancockii. It is found that both 6-BA and KT can up-regulate the endogenous metabolite content and have different effects on aromatic components. Cytokinins significantly affect the aromatic components of A. hancockii, and plant tissue culture is an important means to alter the pattern of aromatic components in aromatic plants.

A concentration-descending washing strategy with methanol for the enhancement of protein imaging in biological tissues by MALDI-MS

Summary: A new method named concentration-descending washing strategy (CDWS) with methanol (MeOH) was successfully developed to enhance the detection and imaging of endogenous proteins in tissues using matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI). Compared to commonly used washing strategies, MeOH-CDWS significantly improved the number and intensity of protein ion signals and reduced protein delocalization, resulting in improved quality of protein images in tissue sections through MALDI-MSI.

In Situ Proteomic Analysis of Herbicide-Resistant Soybean and Hybrid Seeds via Matrix-Assisted Laser Desorption/Ionization-Mass Spectrometry Imaging

Summary: Transgenic soybean, with the largest cultivation area globally, poses ecological risks through gene flow to wild relatives. This study used MALDI-MSI and UPLC-Q-TOF-MS to analyze protein changes in transgenic soybean, wild soybean, and their F2 hybrid. Specific differentially expressed proteins (DEPs) in wild soybean were identified, while F2 seeds exhibited characteristics of both parents. Sucrose synthase and stress response-related DEPs were differentially expressed, potentially contributing to greater adaptability in offspring. MALDI-MSI showed potential as a visual method for transgenic soybean analysis.
