Enrichment Method of Sulfated Glycopeptides by a Sulfate Emerging and Ion Exchange Chromatography

Sulfated glycoproteins are of growing importance for biomarker discovery, as well as for investigating molecular recognition processes. Mass spectrometry (MS) has become a powerful technique for the characterization of glycans and glycoproteins. However, characterization and detection of sulfated glycopeptides by MS is difficult because of the low abundance and low ionization efficiency of these molecules. To overcome this problem, we developed a novel enrichment procedure for sulfated glycopeptides. The procedure consists of anion exchange chromatography and a sulfate emerging (SE) method which controls the net charge of peptides by utilizing limited proteolyzes and modification with acetohydrazide. Using this procedure, we are able to enrich and characterize the sulfated glycopeptides of bovine luteinizing hormone (bLH). Furthermore, we demonstrate the enrichment and detection of sulfated glycopeptides from a complex mixture comprising human serum spiked with bLH at a concentration of 0.1%.

Association between the expression of core 3 synthase and survival outcomes of patients with cholangiocarcinoma

Summary: The study identified potential biomarkers for predicting the prognosis of patients with cholangiocarcinoma (CCA), showing that positive expression of G8-144 was associated with improved prognosis while positive expression of MECA-79 indicated poor prognosis. Both antibodies G8-144 and MECA-79 could be considered as favorable and unfavorable biomarkers for CCA prognosis, respectively, based on their mirror-image staining patterns.

Summary: In this study, a new measurement system was developed to investigate the potential of Wisteria floribunda agglutinin-reactive ceruloplasmin as a serum marker for ovarian clear cell carcinoma (CCC). The specificity and sensitivity of the serum marker measurement were greatly improved, allowing for the accurate diagnosis of CCC. This method can be used for the serodiagnosis of early-stage CCC.

Summary: In this study, the expression of Fut9 and Lewis X in the brain and retina was investigated. Fut9 was found to be expressed in cortical neurons and corticothalamic projection neurons. The loss of Fut9 resulted in a reduction in the number of neurons in the cortex and retina, indicating the significant roles of Fut9 in the differentiation, migration, and maturation of neural precursor cells.

Summary: This study revealed the mechanism of ppGalNAc-T18 ER localization and the function of each structural domain. It was found that the luminal stem region and catalytic domain of ppGalNAc-T18 are essential for ER localization, while the lectin domain and N-glycosylation are not required. The findings provide insights into the subcellular localization mechanism of ppGalNAc-T18 and its role in non-catalytic O-GalNAc glycosylation activity.
