Summary: In this work, an online extraction electrospray ionization (OE-ESI) ion source was developed for real-time analysis of metabolites in vivo without sample pretreatment. The ion source showed excellent salt tolerance and stability, allowing successful mass spectrometry analysis in high salt systems and avoiding the complex desalting process. In a rat liver ischemia-reperfusion model, real-time monitoring of metabolite concentrations revealed varying degrees of change compared to the control group.

Summary: The study reports the development of in vivo subcellular high-resolution mass spectrometry for proteo-metabolomic molecular systems biology in complex tissues. Through precision-translated fabricated microcapillaries, subcellular content of different cell types was analyzed with differential quantitative activities of proteins and metabolites identified. The technology preserved integrity of analyzed cells, neighboring cells, and embryos, with 95% of analyzed embryos developing into sentient tadpoles indistinguishable from wild-type siblings.

Summary: A simple and efficient enrichment strategy based on Amino-functionalized Polymer-Magnetic MicroParticles (NH2-PMMPs) with coral-like porous structures was developed to detect 320 anionic metabolites, including 102 polar phosphate metabolites. Previously unknown polar phosphate metabolites were identified in serum samples using this enrichment method, demonstrating its advantages for mass spectrometric analysis. The high sensitivity of the method enabled the detection of 36 polar anion metabolites from 10 cell equivalent samples.
